RP-HPLC法测定新乡种植红豆杉树皮中的紫杉醇

         

摘要

Objective: Developped a RP-HPLC method for determination of taxol in chinese yew planted in Xinxiang.Method: Agilent TC-C18 Column (250.0 mm×4.6 mm, 5 μm)was used. The mobile phase was ACN-H2O(50:50) at a flow rate of 1 mL/min with the detection wavelength 227 nm. Results:The linear relationship of peak area and injection amount of Taxol was in the range 0.04 μg ~0.134 μg, r=0.999. The average recovery was 99.68 % and RSD was 2.43 %. Conclusion: A reliable and convenient method was firstly established to determine the taxol content in Chinese yew planted in Xinxiang and the result was 0.039 5 % (n=4). It could be used for quality control of Chinese yew planted in Xinxiang.%目的:首次确立了新乡种植红豆杉树皮中紫杉醇的测定方法,方法:反相高效液相色谱法(RP-HPLC),Agilent TC-C18色谱柱(250.0 mmx4.6 mm,5μm);流动相:V(乙腈):V(水)=50:50;流速:1 mL/min;检测波长:227 nm.结果:紫杉醇的线性范围为:0.04μg~1.2μg,r=0.999;平均回收率为99.68%,RSD=2.43%.结论:新乡种植红豆杉树皮中紫杉醇平均含量为0.039 5%(n=4).本法操作简便、准确,重现性好,为新乡种植红豆杉提取紫杉醇的测定及其质量控制的提供了方法依据.
