冬连胶囊调控 AGEs-MAPK通路对糖尿病肾病大鼠肾脏、胰腺保护作用的研究

         

摘要

目的::探讨冬连胶囊通过调控AGEs-MAPK通路对链脲佐菌素( STZ )诱导的糖尿病肾病大鼠肾脏及胰腺的保护作用。方法:雄性SD大鼠60只,随机分为正常组10只,模型组50只。模型组大鼠腹腔注射链脲佐菌素(STZ,45 mg/kg),模型成功的大鼠按体质量随机分为模型组、氯沙坦组、冬连胶囊低、中、高剂量组。分别于灌胃后第1、3、8周收集24 h尿液检测24 h尿蛋白定量。灌胃第8周,处死大鼠取肾组织及胰腺组织,进行病理形态学观察,同时检测组织中MDA、SOD、GSH-Px的含量;腹主动脉取血,检测模型大鼠血液中血糖、HbA1 c﹪、肝肾功、及氧化应激等指标。结果:与模型组比较,冬连胶囊各治疗组可不同程度降低血液中血糖、HbA1 c及肝、肾功水平;冬连胶囊各治疗组可不同程度降低模型大鼠的24 h尿蛋白量,减轻模型大鼠的肾比重;病理结果显示冬连胶囊治疗组对肾脏及胰腺具有一定保护作用;冬连胶囊各治疗组可降低肾组织、胰腺组织中MDA含量,提高组织中SOD、GSH-Px的活性。结论:冬连胶囊可能通过调控AGEs-MAPK信号表达,减轻氧化应激反应,降低血糖及24 h尿蛋白,从而达到保护肾脏及胰腺的作用。%Objective:To discuss the protective effect on kidney and pancreas by Donglian capsules for treating diabetic nephropathy rats by modulating AGEs -MAPK pathway .Methods:Sixty SD male rats were divided into two groups:a normal group with 10 and a model group with 50.The rats in the model group were injected by abdominal Streptozotocin(STZ)(45 mg/kg).After successful models,50 model rats were randomized into five groups:model group , Cozaar group and Donglian low -dose group and Donglian middle -dose group and Donglian high-dose group.Collect 24 h urines and test 24 h urinary protein in 1,3,8 weeks after lavage.In 8 lavage ,kill rats and get out kidney and pancreas tissues ,and then observe the pathological changes ,at the same time test the contents of MDA , SOD GSH -Px of tissues;collect blood in abdominal aorta and detect HbA1c﹪,indexes of liver and kidney function ,and MDA,SOD GSH-Px.Results:Compared with the model group , Donglian capsules treatment groups could reduce 24 h urinary protein quantity of the STZ model rats to different levels , and it could reduce the kidney proportion of model rats and protect kidney of diabetic ne -phropathy model rats;pathological results showed that Donglian capsules had a certain protective effect on the kidney and pancreas;they could reduce MDA ,increase activity of SOD ,GSH-Px in kidney and pancreas tis-sues.Conclusion:Donglian capsules may regulate AGEs -MAPK signal pathway , reduce oxidative stress , low-er blood sugar and 24 h urinary protein ,and then relieve kidney damage so as to protect the kidney and pan-creas,which have a protective effect and delay the progression of diabetic nephropathy .
