骨形态发生蛋白-6对过氧化氢造成的视网膜色素上皮细胞损伤的保护作用

         

摘要

AIM:To investigate the effect of bone morphogenetic protein 6(BMP-6) on ceIIuIar morphoIogy, proIiferation and apoptosis of retinaI pigment epitheIiaI ceIIs(ARPE-19) incubated in hydrogen peroxide( H2 O2 ) . METHODS:ARPE-19 ceIIs were cuItured conventionaIIy and divided into four groups. One group was untreated as bIank group, the other three groups were incubated in 75μm/L H2 O2 , 150ng/mLBMP-6 or75μm/L H2 O2+150ng/mL BMP-6. AII the groups were incubated for 3h, 6h, 9h and 12h. We tested the ceII viabiIitity by MTT. We used fIow cytometry to test the ceII cycIe and ceII apoptosis. RESULTS:H2 O2 significantIy decreased the ceII activity in time-dependent manner. The activity of ceIIs with BMP-6+H2 O2 was higher H2 O2 group, and the differences between the two groups at 3h and 6h were significant (P<0. 05). The observation on ceIIuIar morphoIogy showed that the ceI I number decreased and the ceII detachment after 6h incubated with H2 O2 , whiIe the ceIIs with BMP-6 were Iess ceII detachment and apoptosis. CONCLUSION:BMP-6 has protective effects on RPE ceIIs from oxidative stress in certain extent.%目的::观察骨形态发生蛋白-6( bone morphogenetic protein,BMP-6)对过氧化氢( hydrogen peroxide,H2 O2)作用的人视网膜色素上皮株ARPE-19细胞的细胞形态、增殖以及凋亡的影响。方法:常规培养 ARPE-19细胞,分为正常对照组、75μmol/L H2 O2及150ng/mL BMP-6、75μmol/L H2 O2+150 ng/mL BMP-6环境中培养3、6、9、12 h后, MTT比色法检测细胞的活性;流式细胞仪检测细胞的周期及凋亡变化。结果:H2 O2作用组的细胞活性随着作用时间的延长逐步降低;而BMP-6+H2 O2组的细胞活性则较H2 O2组高,在3h及6h时两组相比差异有统计学意义(P<0.05);细胞形态观察发现:H2 O2培养6h后,细胞数量减少,细胞发生脱落,而添加 BMP-6后细胞脱落及凋亡均要明显减少。结论:BMP-6可以一定程度上保护视网膜色素上皮受到氧化应激的损伤。
