端粒酶催化亚单位TERT在实验性外伤性PVR视网膜前膜的动态表达

         

摘要

AIM: To investigate whether telomerase catalytic sub-unit (TERT) takes part in the regulation of cell proliferation in epiretinal membranes in traumatic proliferative vitreoretinopathy (PVR) and its dynamic change.METHODS: By using S-P technique, immunocytochemi cal staining and HE staining of epiretinal membranes of traumatic PVR were conducted. The results were analyzed by using the computerized image analysis system.RESULTS: At 7, 14, 21, and 28 days after operation,the TERT positive rate was (4.5±2.7)%, (14.2±4.6)%,(13.0±4.8)% and (9.2±2.3)% respectively and the average 4 is 0.0690±0.0213, 0.0912±0.0125, 0.0825±0.0278and 0.0744±0.0159 respectively. The values at day 7were significantly different from those at either day 14 or day 21. The positive rate at day 28 was significantly lower than that at either day 14 or day 21.CONCLUSION: The activation of telomerase may beone of the mechanisms that initiate cell proliferation in epiretinal membranes in PVR. The expression of TERT fluctuates with the PVR course.%目的:探讨端粒酶催化亚单位(TERT)是否参与了实验性外伤性增殖性玻璃体视网膜病变(PVR)视网膜前膜的细胞增殖调控及其随病程的动态变化.方法:对外伤性PVR大鼠模型视网膜前膜做S-P 法TERT免疫组织化学染色和HE染色,染色结果行计算机图像分析.结果:术后7,14,21,28d视网膜前膜阳性细胞率出现先升高后略降,分别为(4.5±2.7)%,(14.2±4.6)%,(13.0±4.8)%,(9.2±2.3)%,平均A分别为0.0690±0.0213,0.0912±0.0125,0.0825±0.0278,0.0744±0.0159,7d组与14d组和21d组比较有显著性差异,28d组较前2组阳性率下降,差别有显著性意义.结论:端粒酶的激活可能是启动PVR视网膜前膜细胞增殖的机制之一,TERT的表达随病程而出现波动.
