乳腺癌EGFL7及β-catenin蛋白检测的临床意义

         

摘要

目的:检测并探讨类表皮生长因子域-7(epidermal growth factor-like domain 7,EGFL7)和β-链蛋白(β-catenin)在乳腺癌患者病变组织及血清中的表达水平及其与乳腺癌转移之间的关系.方法:分别采用免疫组织化学EnVision二步法检测EGFL7及β-catenin蛋白在93例乳腺癌组织(其中淋巴结转移病例62例,远处转移病例10例)与54例乳腺纤维腺瘤和腺病组织中的表达,ELISA双抗体夹心法检测患者血清中EGFL7蛋白水平.结果:EGFL7蛋白在乳腺癌组织中的表达高于乳腺纤维腺瘤和腺病组织(P<0.05),EGFL7蛋白表达水平与乳腺癌淋巴结和远处转移呈正相关(P<0.05).与伴淋巴结转移的乳腺癌病例相比,伴远处转移的乳腺癌病例EGFL7蛋白表达水平差异无统计学意义(P>0.05).乳腺癌组织β-catenin蛋白表达水平高于乳腺良性病变组织(P<0.05),伴有淋巴结和远处转移的乳腺癌组织β-catenin蛋白表达水平高于无淋巴结转移的乳腺癌组织(P<0.05),而伴远处转移的乳腺癌病例的β-catenin蛋白表达水平与伴淋巴结转移的乳腺癌病例比较,差异无统计学意义(P>0.05).发生淋巴结和远处转移的乳腺癌患者血清中EGFL7蛋白水平高于无转移的乳腺癌及乳腺纤维腺瘤和腺病组织(P<0.05),其中伴远处转移的乳腺癌病例血清EGFL7蛋白水平与只伴淋巴结转移的乳腺癌病例比较差异无统计学意义(P>0.05).结论:EGFL7及β-catenin蛋白检测对评估乳腺癌转移风险具有一定的临床参考价值.%Objective: To detect and investigate the epidermal growth factor-like domain 7 (EGFL7) and beta chain protein (β-catenin) expression level in tissue and serum in patients with breast cancer and its relationship with metastasis of breast cancer. Methods: EGFL7 and β-catenin protein were detected by immunohistochemistry EnVision two steps method in 93 cases of breast cancer (including 62 cases with lymph node metastasis and 10 cases with distant metastasis) and 54 cases of breast fibroadenoma and adenosis, EGFL7 protein in serum was detected by ELISA double antibody sandwich method in these cases. Results: The protein expression of EGFL7 in breast cancer was higher than that in breast fibroadenoma and adenosis (P<0.05), the protein expression level of EGFL7 was positive correlation in with lymph node and distant metastasis in breast cancer patients (P<0.05), but there was no statistical difference between EGFL7 protein expression level in breast cancer with distant metastasis and breast cancer cases with lymph node metastasis (P>0.05). The β-catenin protein expression in breast cancer tissue was higher than that of benign breast lesions (P<0.05). The β-catenin protein expression in breast cancer with lymph node and distant metastasis than those without lymph node metastasis in breast cancer tissues (P<0.05), but there was no significant difference between the β-catenin protein expression level in breast cancer patients with distant metastasis and breast cancer cases with lymph node metastasis (P>0.05). The EGFL7 protein expression in serum of breast cancer patients with lymph node and distant metastasis was higher than that in non-metastatic breast cancer and breast fibroadenoma and adenosis (P<0.05), but EGFL7 protein levels were no significant difference in serum of breast cancer patients with distant metastasis compared with the breast cancer with lymph node metastasis (P>0.05). Conclusion: The detection of EGFL7 and β-catenin protein have a certain clinical reference value for evaluating the risk of metastasis of breast cancer.
