周期性张应力对牙周膜成纤维细胞增殖的影响

         

摘要

目的 探讨周期性张应力对人牙周膜成纤维细胞(hPDLF)增殖的影响及其机制。方法采用多通道细胞牵张应力加载系统,以hPDLF为对象构建细胞体外培养-力学刺激模型。加力组分别给予1、2、4、6、12和24h的力学刺激,刺激幅度10%、频率为0.1 Hz。以静态组为对照组。应用细胞计数试剂-8(CCK-8)检测hPDLF增殖情况,RT-PCR检测增殖性细胞核抗原(PCNA)和核因子-κB(NF-κB)的表达;以NF-κB特异性抑制剂吡咯烷二硫代氨甲酸盐(PDTC)预处理细胞作为对照组,检测hPDLF增殖情况。结果加力组细胞在加力1、2h增殖下降,6 h开始增殖,12h增殖达高峰,24h增殖受到明显的抑制;PDTC抑制细胞增殖以及pcna和nf-κBmRNA的表达。结论在一定的时间范围内,周期性张应力能促hPDLF增殖:随着时间的延长,细胞增殖受抑制;NF-κB信号传导通路在周期性张应力介导的hPDLF增殖中发挥着重要的作用。%Objective To study the cyclic stretch on human periodontal ligament fibroblasts (hPDLF) prliferation and its mechanism. Methods In vitro culture-tensile stimulate models of hPDLF were established by using a multi-passage load adding system. Cyclic stretch was applied on the fibroblasts for 1, 2, 4, 6, 12 and 24 hours, respectively, the load was set for 10% surface elongation, with frequency 0.1 Hz. At the same time, the normal control group was established by static group. The cell promote was determined by cell counting kit-8 (CCK-8) method; and the expression of mRNA was detected on proliferating cell nuclear antigen (PCNA) and nuclear fa-ctor-KB (NF-kB) by RT-PCR technology. Results Compared with static group, the proliferation activity of hPDLF was decreased in 1, 2 h; hPDLF began to proliferate in 6 h, cell proliferation reached the peak in 12 h; the proliferation of hPDLF were inhibited obviously. Pyrrolidine dithiocarbamate (PDTC) inhibited cell proliferation and inhibited mRNA expression of PCNA and NF-kB; and the differences between the two groups were significant. Conclusion In a certain time, within the scope of cyclic stretch can promote the in vitro hPDLF proliferation; as time went by, the hPDLF was obvious inhibition. NF-kB signaling pathway play an important role during cyclic stretch-induced hPDLF proliferation.
