基于拉曼和试剂技术的蛋白质定量分析进展

         

摘要

Reagent- and Raman-based protein assay techniques have been developed,and these are routinely used by all the laboratories involved in protein research. In reagent-based protein assay,an unknown concentration of proteins can be determined using a standard curve with known concentrations of a purified reference protein. Most reagent-based protein quantitation techniques such as the Lowry,Bradford,and Bicinchoninic acid( BCA)assays are widely used in quantitative protein research. These assays can be divided into two types:( i)dye-based assays( Bradford assay)and( ii)copper-based assays( Lowry and BCA protein assays). Most reagents,which are used for colorimetric protein assay,possess high Raman scattering cross section. Recently,Raman spectroscopy has attracted considerable interest in reagent-based protein assay owing its potential applications in sensitive and selective protein identification techniques.%基于拉曼技术和试剂技术的蛋白质研究是近些年发展的一种蛋白质定量分析手段。试剂技术是蛋白质实验室研究的最基本手段,它可以利用吸光强度或颜色深浅来判断未知体系中蛋白质的含量,进而建立一套标准来对蛋白质进行定量鉴定。目前,Folin-酚试剂法( Lowry)、考马斯亮蓝法( Bradford)和BCA法是主要用于蛋白质定量分析的技术。而大多数试剂具有显色效应,同时具有高的拉曼散射截面。因此将拉曼光谱法引入到蛋白质定量检验中具有检测灵敏度更高,分析速度快等优势。
