不同方法对柠条锦鸡儿中总黄酮含量测定的影响

         

摘要

[Objective]To compare the effect of different extraction methods on content of total flavones from Caragana korshinskii and optimize the most suitable method. [Method] Ultrasonic-water bath extraction, liquid nitrogen-ethanol extraction, microwave extraction method were adopted to extract total flavones from C. korshinskii, and measured by UV spectrophotometer at 510 nm. [Result] The total flavone content of leaves is 3.88% and the root skin is 0. 83% by the ultrasonic-water bath extraction; the total flavone content of leaves is 3.14% and the root skin is 0.44% by the microwave extraction; the total flavone content of leaves is 2.39% and the root skin is 0.14% by the liquid nitrogen-ethanol extraction. Total flavone content is the highest by ultrasonic-water bath extraction, and the microwave extraction is the second and liquid nitrogen-ethanol extraction is the minimum. [ Conclusion] The study provides basis for development and utilization of flavones, lays a foundation for comprehensive utilization of C. korshinskii.%[目的]比较不同方法提取柠条锦鸡儿总黄酮的含量,以优选出柠条锦鸡儿总黄酮的最佳方法.[方法]分别采用超声波-水浴提取法、液氮-乙醇提取法、微波提取法提取柠条锦鸡儿总黄酮,并用紫外分光光度计在波长510 nm处测定总黄酮的含量.[结果]超声波-水浴提取法测得柠条锦鸡儿叶总黄酮含量为3.88%,根皮为0.83%;微波提取法测得叶中总黄酮含量为3.14%,根皮为0.44%;液氮-乙醇提取法测得叶中总黄酮含量为2.39%,根皮为0.14%.超声波-水浴提取法提取总黄酮含量最高,微波提取法次之,液氮-乙醇提取法最低.[结论]研究可为柠条锦鸡儿中黄酮类物质开发利用提供依据,为柠条锦鸡儿的综合开发利用奠定基础.
