[目的]建立HPLC法同时测定大黄中芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚含量,并对不同产地的大黄样品进行测定.[方法]采用依利特ODS2 C18柱(200 mm × 4.6 mm,5μm),流动相为无水甲醇-浓度0.1%磷酸(75∶25,v/v),流速为1 ml/min,检测波长为254nm,柱温25℃.[结果]芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚的线性回归方程分别为:Y=1.96X-0..25,相关系数R =0.999 94;Y=2.45X+0.18,相关系数R=0.999 94;Y=1.91X +0.05,相关系数R=0.999 90;Y=1.25 X-0.06,相关系数R=0.999 95;Y=0.96 X-0.20,相关系数R=0.999 98;平均回收率分别为99.39%、98.84%、98.94%、99.11%和98.37%.[结论]该方法简单、快速、重现性好、结果准确,可用于大黄药材品质评价与质量检测.%[Objective] The establishment of HPLC method for simultaneous determination of aloe emodin in rhubarb,rhein,emodin,chrysophanol physcion,content,and different places in rhubarb samples.[Method] ODS2 C18 column (200 mm ×4.6 mm,5 μm),mobilephaseof methanol:0.1% phosphoric acid =75∶25 (V/V),the flow rate was 1 ml/min,the detection wavelength was 254 nm,column temperature was 25 ℃.[Result] The linear regression equation of aloe-emodin; rhein; emodin; chrysophanol ; physcion are Y =1.96X-0.25,correlation coefficient R =0.999 94 ; Y =2.45 X + 0.18,R =0.999 94; Y =1.91 X + 0.05,R =0.999 90 ; Y =1.25 X-0.06,R =0.999 95 ; Y =0.96 X-0.20,R =0.999 98 ; the average recoveries were 99.39%,98.84%,98.94%,99.11%,98.37%.[Conclusion] The method is simple,rapid,reproducible,accurate results,and can be used for quality evaluation and quality test of rhubarb.
