广西巴马小型猪PPARγ-2基因多态性与2型糖尿病易感性的相关性

         

摘要

[目的]探讨PPARγ-2基因多态性与广西巴马小型猪2型糖尿病易感性的关系.[方法]利用高糖高脂日粮饲喂24头广西巴马小型猪,利用PCR方法扩增PPARγ-2基因外显子2部分序列,测定其空腹血糖和胰岛素(INS)含量并进行糖耐量试验.[结果]克隆的PPARγ-2基因外显子2部分序列存在1处SNP位点(19813A/G),有AG(7头)和AA(17头)2种基因型.AG型广西巴马小型猪的空腹胰岛素含量和糖耐量试验中60 min血糖值显著高于AA型(P<0.05),而AG型广西巴马小型猪2型糖尿病发生率(71.4%)明显高于AA型(5.9%).[结论]广西巴马小型猪PPARγ-2基因外显子2的19813A/G多态性与2型糖尿病易感性有关.%[Objective] The research aimed to discuss the relationship between the genetic polymorphism of PPARγ-2 and the susceptibility of type 2 diabetes mellitus(T2DM)in Guangxi Bama mini-pig. [Method] 24 Guangxi Bama mini-pigs were fed with high-fat and high-sucrose diet and partial sequences of exon 2 of PPAR7-2 gene were amplified by PCR method. The contents of blood glucose and insulin in Guangxi Bama mini-pig under fasting conditions were determined and the glucose tolerance test( GTT) was also carried out. [Result] There was one SNP site (19813A→G)in partial sequence of exon 2 of the cloned PPARγ-2 gene. And two genotypes, AA(7 pigs) and AG(17 pigs)were detected. The fasting blood insulin content and 60 min blood glucose level in GTT in AG-genotype Guangxi Bama mini-pig were significantly higher than that of AA genolype( P < 0.05 ). The incidence of T2DM in AG-genotype Guangxi Bama mini-pig (71.4% ) was obviously higher than that of AA genotype (5.9% ). [Conclusion] The polymorphism of 19813A→G in exon 2 of PPARγ-2 gene was related with the susceptibility of T2DM in Guangxi Bama mini-pig.
