[Objective] To study the properties of thermostable xylanase. [Method] The optimal temperature, pH of producing thermostable xylanase by Rosetta(DE3)/pET28a-xynB64, thermal stability, pH stability were investigated, as well as the effects of different metal ions on enzyme activity. [Result] The optimal reaction temperature and pH are 110 ℃ and 6.6 with extensive stability range of pH and good thermal stability. High concentration of Mn2+ , Mg2+ , Ca have activation effect on enzyme, while Cu2+ , Zn2+ and low concentration Mn2+ have inhibition effect on the enzyme. [Conclusion] The study provides reference basis for extensive application of thermostable xylanase.%[目的]研究嗜热木聚糖酶的酶学性质.[方法]考察了基因工程菌Rosetta(DE3)/pET28a-xynB64所产嗜热木聚糖酶反应的最适温度、最适pH,嗜热木聚糖酶的热稳定性、pH稳定性,以及不同金属离子对酶活力的影响.[结果]嗜热木聚糖酶的最适反应温度为110℃左右,最适pH为6.6,具有较广的pH稳定范围和非常好的热稳定性,高浓度的Mn2+、Mg2+、Ca2+对该酶有激活作用,Cu2+、Zn2+以及低浓度的Mn2+对该酶有抑制作用.[结论]研究可为嗜热木聚糖酶的广泛应用提供参考依据.
