阳离子A对内毒素损伤大鼠肝脏中HSP70表达的影响

         

摘要

[ Objective ] The aim was to study the effect of cation A on expression of HSP70 in tat liver injured by endotoxin. [ Method ] Fortyeight SD rats were randomly divided into two groups: the control group and CA + ET treament group. The rats in eontrol group were injected with 5 mg/kg ET,and the rats in CA +ET group first were injected with I g/kg CA,then with 5 rag/kg ET after 2 minutes. After three,four,eight and twelve hours liver samples were taken, and detected with HE staining and immunohistochemieal staining. [ Result ] The pathologic change of the liver obviously increases after endotoxin attacks 3 h,4 h reaches peak value ,drop after 8 h gradually. The liver cell lesions of CA + ET group was slightly reduced at the same time compared with control group; HSP70 of the two groups in the integrated optical density in 3,8,12 h was significantly ( P < 0. 0l ) ,4 h significant difference ( p < 0.05 ), Integrated optical density of HSP70 declined with time in control group,while in CA + ET group,the integrated optical density of HSP70 in 3,4,8 h these three time points decreased a modest increase in 12 h. [ Conclusion] CA could upregulate the expression of HSP70 in the liver significantly,which can protect rat liver from injury of iraduced-Endotoxin.%[目的]运用免疫组织化学技术研究阳离子A(CA)对HSP70在内毒素(ET)损伤大鼠肝脏中表达的影响.[方法]48只SD大鼠随机分为2组:对照组和CA+ET组.每组各24只.对照组大鼠分别经尾静脉注射ET 5 mr/kg(BW),CA+ET组尾静脉注射1 g/kg(BW)CA溶液,之后2 min内尾静脉注射ET 5 mg/kg BW.2组分别在3、4、8、12 h采集肝脏作为检测样本,应用HE染色和免疫组织化学染色技术进行检测.[结果]肝细胞病理变化在ET攻击后3和4 h时明显增多.4 h达到峰值,而在8和12 h时逐渐下降.CA+ET保护组肝细胞的细胞病变较ET组同时间段有所减轻;2组HSP70的积分光密度在3、8、12 h差异极显著(P<0.01),4 h差异显著(P<0.05),且对照组HSP70积分先密度随时间的延长而下降,而CA+ET组HSP70的积分光密度在3、4、8 h这3个时间点呈下降趋势,在12 h小幅度上升.[结论]CA能显著上调HSP70在肝脏的表达,从而对由ET诱导的肝损伤产生保护效应.
