白玉银桦组培快繁技术研究

         

摘要

[Objective] The research aimed to study the tissue culture of G. baileyana,to improve its reproduction rate and the rooting rate and establish an economic and high effective technology system of rapid propagation of G. baileyana. [ Method] The suitable inducing medium,multiplication medium and rooting medium of semi-ligmified branches of G. baileyana were studied. [ Result ] The optimal inducing medium was MS + 6-BA 1. 0 mg/L + AA 0.1 mg/L + sucrose 30 g/L + agar powder 6 g/L. The optimal multiplication medium was MS + 6-BA 1. 0 mg∕L + KT 0.2 mg/L + NAA 0. 3 mg/L + sucrose 30g/L + agar powder 6 g/L,and the highest multiplication rate was 3.6. The optimal rooting medium was 1/2MS + NAA 0.3 mg/L + sucrose 20 g/L + agar powder 6 g/L,and the rooting rate reached 90 % and the average number of root was 3.0 when culturing on them. [ Conclusion ] The study sets up a practical and effective measure and way for solving the contradiction at present between the supply and requirement of G. baileyana seedling,has important theoretical value and realism significance.%[目的]研究白玉银桦(Grevillea baileyana)的组培快繁技术,以提高繁殖速率和生根率,建立一套高效的再生体系.[方法]以白玉银桦半木质化枝条为外植体,对适宜的丛芽诱导、增殖、生根的培养基进行了系统研究.[结果]最适启动培养基为:MS+6-BA 1.0mg/L+NAA0.1 mg/L+蔗糖30 g/L+琼脂粉6 g/L;最佳增殖培养基为:MS+6-BA1.0 mg/L+KT 0.2 mg/L+NAA 0.3 mg/L+蔗糖30g/L+琼脂粉6 g/L,增殖系数达3.6以上;适宜的生根培养基为:1/2MS+NAA 0.3 mg/L+蔗糖20 g/L+琼脂粉6 g/L,生根率达到90%以上,生根苗平均根数3.0条.[结论]该研究为解决目前白玉银桦种苗的供需矛盾,开辟了一条切实有效的方法和途径,具有重要的理论价值和现实意义.
