费约果外植体消毒处理方法

         

摘要

[目的]获得无菌、生长正常的费约果外植体,从而进行组培快繁.[方法]通过选用费约果不同组织部位作为外植体,采用不同消毒方法及不同预培养的培养基来进行费约果的外植体制备试验,从中筛选出适合的条件.[结果]采用费约果新梢为外植体,用0.1%HgCl2消毒4 min后,在含有羧苄青霉素、硫酸链霉素附加DTT,Vc的1/2MS培养基上预培养21 d获得的外植体消毒效果最好.[结论]获得了生长健壮的费约果外植体,为其快速整殖提供了技术依据.%[ Objective] The research aimed to provide the scientific basis for the development of the medicine and functional food with Cucurbita pepo L polysaccharide. [ Method ] Crude Cucurbita pepo L polysaccharide was extracted from Cucurbita pepo L by hot-water extraction. The content of polysaccharides was measured by anthrone-sulfuric acid method. · OH- was produced by Fenton reaction,and · O2- was produced by the oxidation of pyrogallic acid. [ Result ] When the concentration of Cucurbita pepo L polysaccharide was 2 mg/ml in 37℃ ,and the alcohol quantity was 4 times the volume of ASPs filtrate, the elimination rate of the production to · OH- reached 37. 1 %. When the concentration of Cucurbita pepo L polysaccharide was 2 mg/ml in 25 ℃ , and the quantity alcohol was 4 times the volume of ASPs filtrate, the elimination rate of the production to · OH reached 23.3%. [Conclusion] Cucurbita pepo L polysaccharide had the significant scavenging effect on O2- and OH-.
