通过蛋白质序列比对探讨细菌的热适应机制

         

摘要

[目的]从蛋白的一级序列特征出发探索原核生物的热适应机制.[方法]从526种原核生物的蛋白序列中识别出同源蛋白,并对其进行了综合分析.[结果]找到了存在于超嗜热、嗜热细菌的同源蛋白组( HT-only) 95种和仅存在于常温细菌中的同源蛋白组(M-only)1 659种,发现了氨基酸组成与细菌最适生长温度存在显著的相关性.结合在超嗜热、嗜热、常温和嗜冷细菌中都有分布的同源蛋白组(Top-90),通过分析得出HT-only和Top-90组的同源蛋白簇非常相似,而M-only组与其他2组有所不同.[结论]从原核生物全蛋白序列的角度,证实了氨基酸组成与热稳定性存在的显著相关性,验证了先前学者的关于蛋白氨基酸组成与嗜热机制相关性的研究结论.%[Objective] The therroo-adaptation mechanism of prokaryote was researched from the analysis of (he property of first-level protein se quence. Combined with ine homologous protein group6(Top-90) existing in ultra-thermoph ilic,thermophilic,mesophilic bacieria and psychrophilic bacteria and the analysis,it can be seen that the cluster of nrthologous group(COG) in the HT-only group and the Top-90 gmup was very similar while in the M-only group,different from Ihe other two groups. [Conclusion]It was veri fied that there was a significant correlation between the amino acid composition and thermal stability and also the conclusion of previous scholars on the amino acid composition associated with ihermophilic mechanism from the perspective of whole prokaryolic protein sequence.
