应用电导率仪测定微生物生物量的研究

         

摘要

[Objective] The research aimed to study the determination methods of the biomass of bacteria. [ Method] Liquid bacteria culture was centrifuged and Synechocystis sp. PCC 6803 were collected. After washing by deionized water, the cells of collected Synechocystis sp. PCC 6803 were broken by ultrasonic wave. Then the conductivity of different density of the sonicated bacteria was determined using conductometer. At the same time, the dry weight and light absorption of different density of bacteria were measured. [ Result] The results showed that there is a perfect linear relationship between the cell conductivity and its dry weight, correlation coefficient r = 0.9994; the linear relationship between dry weight and optical density of Synechocystis sp. PCC 6803 is also good, correlation coefficient r = 0.998. [ Conclusion] The optical density method and conductivity method are both good methods in the determination of the biomass of Synechocystis sp. PCC 6803. Comparing with optical density method, conductivity method is also convenient and accurate.%[目的]研究微生物生物量的测定方法.[方法]离心收集蓝细菌,用去离子水清洗后,超声破碎蓝细菌,将破碎后的蓝细菌用去离子水稀释至不同浓度,测定不同浓度破碎后的蓝细菌菌液的电导率值.同时测定蓝细菌的细胞干重及吸光度值.[结果]完全破碎后的蓝细菌菌液的电导率值与细胞干重存在良好的线性关系,线性系数r =0.999 4;蓝细菌的吸光度值与细胞干重也存在良好的线性关系,线性系数r=0.998.[结论]电导率法和吸光度法均能很好地反映蓝细菌的生物量,电导率法的方便程度和可靠性均可以与吸光度法相比较.
