目的 探讨Rho激酶抑制剂盐酸法舒地尔在大鼠肺缺血再灌注损伤中的保护作用及机制.方法 将24只大鼠随机分为3组,假手术组(sham组)、缺血再灌注组(IR组)、缺血再灌注+Rho激酶抑制剂组,缺血1小时再灌注2小时后,获取左肺标本,对三组肺组织进行病理检查,同时检测三组肺组织湿干重比、肺组织肿瘤坏死因子(TNF-α)/白细胞介素(IL-6)、髓过氧化物酶(myeloperoxidase,MPO)的水平.结果 经Rho激酶抑制剂预处理后的肺组织病理损伤较IR组轻,同时肺组织湿干重比、TNF-α/IL-6、MPO的水平也较IR组低,所有差异均有统计学意义,P<0.05.结论 Rho激酶抑制剂通过减少炎性细胞因子的表达及氧自由基的产生在肺缺血再灌注损伤中发挥保护作用.%Objective To investigate the protective effect and mechanism of Rho-kinase inhibitor fasudil on lung ischemia-reperfusion injury in rats. Methods 24 rats were randomly divided into three groups: the sham group,the ischemia-reperfusion group(the IR group),and the ischemia reperfusion + Rho kinase inhibitor group. After 1-hour ischemia and 2-hours reperfusion,wet/ dry weight ratio,TNF-α/ IL-6 and MPO were measured in the lung tissues of the three groups. Results The lung tissues pathological damages after pretreatment with Rho kinase inhibitor decreased more obviously than that with IR,and the wet /dry weight ratio,TNF-α / IL-6 and MPO were al-so decreased more obviously in the Rho group than in the IR group (P<0.05). Conclusion Rho-kinase inhibitor plays a protective role in the procedure of lung ischemia reperfusion injury by reducing the expression of inflammatory cytokines and the production of oxygen free radicals.
