水稻中介体亚基OsMed6的表达及进化分析

         

摘要

中介体复合物是真核生物RNA聚合酶Ⅱ(PolⅡ)通用转录装置的重要组成部分,是基因特异性转录因子与启动子信息传递的桥梁.本研究对水稻(Oryza sativa)中一个假定的中介体亚基OsMed6的表达和进化进行了分析.通过整合分析不同来源的基因芯片数据,发现OsMed6在各个组织(器官)里都有表达.构建了OsMed6与增强型黄色荧光蛋白融合的表达载体,通过农杆菌介导在烟草叶片中进行瞬时表达,表明OsMed6定位在细胞核中.通过多序列联配和进化分析,发现OsMed6与其他真核生物(尤其是植物)的同源蛋白具有很高的序列保守性.%Mediator is a large multiprotein complex required for transcriptional activation in vivo, which provides an interface for activator and repressor proteins to transmit information from regulatory DNA elements to core promoters. In this study, the expression and evolution of 0sMed6, a putative subunit of rice mediator, was analyzed. Joint analysis of microarray data from different sources indicated that OsMed6 is constitutively expressed in both vegetative and floral tissues. Agrobacteriwn-mediated transient expression of the fused protein of OsMed6 and eYFP in tobacco leaves suggested that OsMed6 is specifically located in the nuclear. Multiple a-lignment and phylogenetic analysis showed that 0sMed6 has a high sequence conservation with the homologs from other eukaryotes ( especially other plants ).
