兖州卷柏提取物对CCl4致小鼠肝损伤的保护作用

         

摘要

目的:研究兖州卷柏提取物对四氯化碳(CCl4)引起肝损伤小鼠的保护作用。方法48只KM 小鼠随机分成6组。Ⅰ组和Ⅱ组为正常对照组和肝损伤模型组,分别每天一次性灌胃20 mL/kg生理盐水;Ⅲ组每天一次性灌胃80 mg/kg水飞蓟素;Ⅳ~Ⅵ组分别每天一次性灌胃40,80,120 mg/kg兖州卷柏提取物。各组连续给药7 d。末次给药2 h后,除正常对照组外,其他各组一次性腹腔注射10 mL/kg的0.2% CCL4。禁食不禁水18 h后,称质量;摘眼球取血,分别测定血清中谷丙转氨酶(ALT)和谷草转氨酶(AST)活性;颈椎脱臼法处死小鼠,取小鼠肝脏并称质量,另取小鼠肝脏右叶的相同部位,用10%甲醛固定,进行石蜡切片和 H‐E染色,检测肝组织病理学变化;取肝脏组织匀浆,测定总抗氧化(T‐AOC)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT )活性。结果兖州卷柏提取物各剂量组ALT和AST活性均显著低于模型组(P<0.01),且趋于正常小鼠水平;与模型组比较,兖州卷柏提取物各剂量组均能显著提高肝脏T‐AOC和CAT活性(P<0.01)。除低剂量组外,其他实验组与模型组之间SOD活性均存在显著差异(P<0.01);兖州卷柏提取物能减轻肝组织损伤程度。结论兖州卷柏提取物对C C l4致小鼠急性肝损伤具有保护作用。%Objective To evaluate the hepatoprotective effect of ethanol extract from Selaginella involvens (EESI) on carbon tetrachloride (CCl4 )‐induced hepatic damaged mice . Methods The animals were randomly divided into six groups ,and treated as follows :GroupⅠ and GroupⅡ were normal control group and model group ,and they were both given 20 mL/kg normal saline . Group Ⅲ were given 80 mg/kg silymarin . Group Ⅳ ,Ⅴ and Ⅵ were given EESI with the dose of 40 ,80 ,120 mg/kg respec‐tively . All groups were given medicines or normal saline via intragastric administration once a day lasting for 7 days . 2 h after the last intragastric administration ,all animals were given 0 .2% CCl4 with the dose of 10 mL/kg except normal control group . After 18 h ,all animals were weighted and blood sample were collected by eye bleeding method . The activities of alanine transaminase (ALT) and aspartate transami‐nase(AST) of blood sample were evaluated . Following the sacrifice for all the animals ,the livers were dissected . Thereafter ,the activities of total antioxidant capacity (T‐AOC) ,catalase(CAT) and superox‐ide dismutase(SOD) of liver supernatant were tested . The livers were fixed in 10% formaldehyde at room temperature for histological analysis . Results Administration of EESI significantly reduced the level of ALT ,AST(P<0 .01) . Compared with model group ,administration of EESI could significantly enhance the level of T‐AOC ,CAT(P<0 .01) . Groups other than the low dose group ,significant differences in SOD activity existed between model group and EESI administration groups(P<0 .01) . Administration of EESI could alleviate liver injury in sinusoidal spaces in CCl4‐induced hepatic mice . Conclusion EESI may protect hepatic injury in CCl4‐induced mice .
