目的:探讨小窝蛋白2(CAV2)在口腔癌患者中的表达及其临床意义.方法:首先用GCBI数据平台下载口腔癌mRNA芯片数据(GSE25099、GSE42743、GSE9844和GSE30784),再利用Stata软件实现诊断准确性试验Meta分析来评估CAV2在口腔癌中的表达情况;利用Oncomine数据库验证CAV2在口腔癌中的表达情况;采用免疫组织化学染色方法验证CAV2在口腔癌组织中的表达;利用TCGA数据库下载口腔癌和口腔正常组织的临床资料和CAV2 mRNA表达水平数据,再次验证CAV2的表达并分析其与患者临床参数的关系,分析CAV2在肿瘤中的诊断价值和患者的生存情况.结果:与正常对照组织相比,生物信息学分析以及免疫组织化学染色结果均提示CAV2在口腔癌中表达上调,差异具有统计学意义(P＜0.05).CAV2的表达与患者的年龄、性别、肿瘤原发灶、组织分级和TNM临床分期无相关性(P＞0.05).CAV2诊断口腔癌的ROC曲线下面积为o.855(95％ CI:0.806～0.905,P＜0.0001),灵敏度为71.3％,特异度为93.8％(最佳临界值为13.63).高表达CAV2的患者生存时间较低表达患者短(x2=6.457,P-0.011).结论:CAV2表达水平可作为口腔癌诊断和预后评估的指标.%Objective:To investigate the expression of caveolin 2 (CAV2) in oral cancer and its significance.Methods:Gene-Cloud of Biotechnology Information (GCBI) platform was used to download 4cDNA microarray datasets of oral cancer (GSE25099,GSE42743,GSE9844 and GSE30784).Meta-analysis was used to analyze the mRNA level of CAV2 by Stata 12.0 software.Oncomine database was used to verify the expression of CAV2 in oral cancer.The protein expression of CAV2 in oral cancer tissues was detected by immunohistochemical staining.Clinical data of oral cancer and normal oral tissues and their CAV2 mRNA levels were downloaded from TCGA database.The expression of CAV2 in oral cancer was verified.And the association of CAV2 expression and the clinical parameters were analyzed.Results:Bioinformatics analysis and immunohistochemical staining were suggested that CAV2 was up-regulated in oral cancer patients compared with the normal controls (P＜0.05).The expression of CAV2 was not correlated with the age,sex,primary tumor,histological grade and TNM staging of the patients.The area under the ROC curve of CAV2 for the diagnosis of oral cancer was 0.855 (950％ CI:0.806-0.905,P＜0.0001),and the sensitivity and specificity were 71.3％ and 93.8％ with a cut off value of 13.63.The survival time of patients with low expression of CAV2 was longer than that of patients with high CAV2 expression (x2 =6.457,P=0.011).Conclusion:CAV2 could be served as a marker for diagnosis and prognosis evaluation of oral cancer.
