HPLC-MS/MS法测定婴幼儿配方乳粉中胆碱、左旋肉碱、牛磺酸与肌醇

         

摘要

A high performance liquid chromatography-tandem mass spectrometric(HPLC-MS/MS) method was established for the simultaneous determination of 4 kinds of optional components (choline, L-carnitine, taurine and inositol) in infant formula milk powder. After dissolution with warm water, the protein in sample was precipitated with zinc acetate and potassium ferrocyanide. The supernatant was separated on an HSS T3 column, then detected by MS/MS under MRM mode, and quantified by the internal and external standard methods. Under the optimized conditions, the calibration curves were linear in the ranges of 0.01-2.0 mg/L for choline and L-carnitine, and 0.1-2.0 mg/L for taurine and inositol, with correlation coefficients larger than 0.997. The detection limits of the method were 1. 5 mg/kg for choline and L-carnitine, and 15 mg/kg for taurine and inositol, respectively. The mean recoveries were in the range of 87. 5%-102. 4% with relative standard deviations (RS-Ds, n =6) of 3.0%-7.3%. The method, with the advantages of high sensitivity, good purification effect and quantitative accuracy, is applicable for the detection of choline, L-carnitine, taurine and inositol in infant formula milk powder.%建立了一种同时测定婴幼儿配方乳粉中4种可选择成分(胆碱、左旋肉碱、牛磺酸和肌醇)的高效液相色谱-串联三重四极杆质谱(HPLC-MS/MS)分析方法.样品经温水溶解后用亚铁氰化钾和乙酸锌沉淀蛋白,上清液过滤后采用HSST3色谱柱分离,三重四极杆质谱仪检测,胆碱和左旋肉碱使用内标法定量,牛磺酸和肌醇使用外标法定量.在最优化条件下,胆碱和左旋肉碱在0.01~2.0mg/L范围内,牛磺酸和肌醇在0.1~2.0mg/L范围内呈良好的线性关系,相关系数均大于0.997;胆碱和左旋肉碱的检出限均为1.5 mg/ kg,牛磺酸和肌醇的检出限均为15 mg/kg. 4种化合物的回收率为87.5%~102.4%,相对标准偏差(RSD,n =6)为3.0%~7.3%.该方法灵敏度高、净化效果好、定量准确,适用于婴幼儿配方乳粉中胆碱、左旋肉碱、牛磺酸和肌醇的同时快速检测.
