维生素D缺乏饲料联合高果糖糖水喂养诱导小鼠非酒精性脂肪肝炎

         

摘要

目的 探讨维生素D缺乏饲料联合高果糖糖水诱导小鼠非酒精性脂肪肝炎(NASH)发生发展情况及其与炎症、脂肪酸代谢的关系.方法 80只小鼠随机分为4组:标准饲料组(C组),用AIN93基础饲料喂养;高果糖糖水标准饲料组(Hfr组),用20%果糖糖水加AIN93基础饲料喂养;维生素D缺乏饲料组(VDD组),用AIN93缺乏维生素D饲料喂养;缺乏维生素D加高果糖糖水饲料组(Hfr/VDD组),用20%果糖糖水加AIN93缺乏维生素D饲料喂养.喂养7个月后,检测血液生化指标,肝脏组织病理切片染色,检测脂肪酸合成分解相关基因、纤维化和炎症相关基因转录水平.结果 单独Hfr组或VDD组饲料未诱发NASH;Hfr/VDD饲料诱发出NASH,原癌基因c-Myc、谷氨酰胺合成酶在肝脏中表达丰度也升高.结论 Hfr/VDD饲料可诱发NASH,充足维生素D可有效地抑制原癌基因表达;VDD饲料喂养的小鼠肝脏原癌基因c-Myc上调,促进脂肪酸合成,抑制脂肪酸分解,肝脏脂肪堆积.%Objective To investigate into the development of non-alcoholic steatohepatitis and the correlation between inflammation and fatty acid metabolism.Methods A total of C57BL/6J mice were randomly divided into 4 groups with different diets:standard-chow group (C),high fructose drinking group (Hfr),vitamin D deficient group (VDD),high fructose drinking plus Vitamin D deficienct diet group (Hfr/VDD).After feeding of seven months,they were examined for physiological and blood biochemical indexes,liver tissue pathology,fatty acid metabolism,inflammation,and fibrosis-related genes expression;and their gene transcription levels were further discussed regarding NASH development.Results We found that Hfr diet alone or VDD diet could not induce NASH,while Hfr/VDD diet could.But in the Hfr/VDD group,proto-oncogene c-Myc and glutamine synthase protein expression increased.Conclusion Hfr/VDD diet can cause NASH.Under the condition of a standard diet,sufficient VD can effectively inhibit proto-oncogene expression and promote fructose catabolism;On the other hand,VDD diet can induce proto-oncogene c-Myc expression,promote the synthesis of fatty acids and inhibit fatty acid catabolism.
