目的 研究冠状动脉粥样硬化性心脏病(冠心病,CHD)患者的载脂蛋白E(ApoE)基因多态性及其与血清脂质水平之间的关系.方法 采用生物化学方法检测401例CHD(CHD组)的总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL)、高密度脂蛋白胆固醇(HDL)、载脂蛋白A1(ApoA1)、载脂蛋白B(ApoB)、脂蛋白a(LPa),应用基因芯片法检测CHD组患者及对照组(60例同期健康者)的ApoE多态性.结果 CHD组共检出6种基因型,分别为E2/E2、E3/E3、E4/E4、E3/E4、E2/E3、E2/E4,对照组检出E3/E3、E3/E4、E2/E3、E2/E4,与对照组相比,CHD组e4等位基因频率增高(P<0.05);CHD组中不同基因型其血脂水平存在一定差异,与E3/E3相比,E2/E2基因型TC、Lpa水平较低,HDL、ApoA水平较高,E4/E4基因型TC、LDL、Lpa水平较高(P<0.05).结论 ApoE多态性与机体脂质代谢密切相关,e4等位基因可能是CHD发病重要遗传因素之一.%Objective To study the relationship between apolipoprotein E(ApoE)gene poly-morphism and serum lipid levels in patients with coronary heart disease(CHD).Methods Levels of total cholesterol(TC),glycerin(TG),low density lipoprotein cholesterol(LDL),high density lipoprotein cholesterol(HDL),apolipoprotein A1(ApoA1),apolipoprotein B(ApoB)and lipopro-tein a(LPa)were detected using biochemical methods in 401 patients with CHD.In addition, ApoE gene polymorphism was measured using gene-chip method in CHD patients and 60 healthy controls.Results Six genotypes(E2/E2,E3/E3,E4/E4,E3/E4,E2/E3,E2/E4)were found in CHD group,and 4 genotypes(E3/E3,E3/E4,E2/E3,E2/E4)in control group.Compared with control group,the frequency of e4 increased in CHD group(P <0.05).Compared with CHD pa-tients with E3/E3 genotype,levels of TC and Lpa decreased and levels of HDL and ApoA in-creased in patients with E2/E2 genotype,and levels of TC,LDL and Lpa increased in patients with E4/E4 genotype(P <0.05).Conclusion ApoE gene polymorphism is closely related to lipid metabolism.The e4 gene may be one of the important genetic factors for CHD.
