与乳腺癌细胞共培养的人成纤维细胞特异蛋白-1表达上调及意义

         

摘要

Objective To investigate the molecular mechanisms of action of stromal fibroblasts on breast cancer,and to study the expression of fibroblast-specific protein-1(FSP-1)and its im-pact on breast cancer growth.Methods Human breast cancer MDA-MB-231 cells and human en-dometrial stromal fibroblasts(ESF)were co-cultured in Transwell system,or cultured alone.The expression of FSP-1 mRNA was examined in MDA-MB-231 cells and ESF by RT-qPCR.The ex-pression of FSP-1 protein was detected by immunofluorescence flow cytometry.The proliferation of MDA-MB-231 cells was tested using Cell Counting Kit-8.The expression of FSP-1 in breast cancer tissue of tumor-burdened nude mice was analyzed using immunofluorescence confocal laser microscopy,and the effect of FSP-1 on tumor volume was examined.Results The expression of FSP-1 was up-regulated in ESF co-cultured with MDA-MB-231 cells,and the up-regulation of FSP-1 promoted the proliferation of MDA-MB-231 cells.FSP-1 was expressed in MDA-MB-231cells co-cultured with ESF,but not in MDA-MB-231 cells cultured alone.In addition,FSP-1 was highly expressed in the cancer tissue with co-growth of MDA-MB-231 cells and ESF in nude mice,and the high expression of FSP-1 accelerated tumor growth.Conclusion The co-culture with human breast cancer cells can up-regulate FSP-1 expression and promote breast cancer growth.%目的:探索乳腺癌间质成纤维细胞对乳腺癌作用的分子机制,研究与人乳腺癌细胞共培养的人成纤维细胞特异蛋白-l(fibroblast-specific protein-1,FSP-l)的表达及对乳腺癌生长的影响。方法Transwell 方法共培养人乳腺癌细胞株 MDA-MB-231和人成纤维细胞株 ESF;实时 RT-qPCR 检测共培养的 MDA-MB-231和 ESF 细胞 FSP-1 mRNA 水平;免疫荧光流式细胞仪检测共培养 MDA-MB-231和 ESF 细胞 FSP-1蛋白表达水平;Cell Counting Kit-8(CCK-8)检测 MDA-MB-231细胞的增殖;免疫荧光激光共聚焦显微镜观察荷瘤裸鼠乳腺癌组织 FSP-1的表达,检测 FSP-1对荷瘤裸鼠肿瘤体积的影响。结果MDA-MB-231细胞和 ESF 细胞共培养可上调 ESF 细胞表达FSP-1并促进 MDA-MB-231细胞增殖;ESF+MDA-MB-231组 MDA-MB-231细胞表达 FSP-1,但在 MDA-MB-231组未检测到 FSP-1蛋白表达;荷瘤裸鼠 MDA-MB-231细胞和 ESF 细胞共生长的癌组织高表达 FSP-1并促进肿瘤生长。结论与人乳腺癌细胞共培养可上调人成纤维细胞 FSP-1的表达并促进乳腺癌生长。
