目的 研究开肺化痰清热止咳中药金欣口服液对呼吸道合胞病毒(RSV)感染的Hep-2细胞TLR4及该通路下游细胞因子TNF-α表达的影响.方法 制备血行药物成分溶液,作用于RSV感染的Hep-2细胞,通过RT-qPCR、免疫荧光标记结合激光共聚焦技术及ELISA法,分别观察分析细胞TLR4 mRNA、蛋白和相关细胞因子的表达水平,分析药物作用机制.结果 与病毒对照组比较,金欣口服液作用于RSV感染的Hep-2细胞后,TLR4 mRNA及蛋白表达显著降低,TNF-α表达显著降低,细胞病变程度减轻.结论 金欣口服液含药血清可以通过下调TLR4和TNF-α表达水平减轻感染RSV后Hep-2细胞的病变程度.%OBJECTIVE To study the influence of Jinxin Oral Liquid (JXOL),a prescription of Chinese medicine which can stop coughing by clearing fever, moistening lung and dissipate phlegm, on the expression of the receptor as TLR4 and the downstream cytokine as TNF-α in respiratory syncyntial virus (RSV) infected Hep-2 cells. METHODS The serum contained JXOL were prepared and RSV infected Hep-2 cells were treated with it. Then cells and supernate were collected respectively and the expression level of Toll-like 4(TLR4) mRNA and protein as well as and the relative cytokines TNF-α were detected by RT-qPCR, Immunofluorescence combined confocal microscopy and ELISA. RESULTS Compared with the control group just treated with RSV, the expression of TLR4 mRNA and protein as well as TNF-α were reduced and the pathological changes were relieved obviously in RSV infected Hep-2 after treated with serum contained JXOL. CONCLUSION The serum contained JXOL can relieve the pathological changes of RSV infected Hep-2 cells by down-regulating the expression of TLR4 and TNF-α.
