卤米松对角质形成细胞维生素D受体表达的影响

         

摘要

ObjectiveTo investigate the effects of halometasone on the expression of vitamin D receptor(VDR) in human keratinocytes so as to evaluate the potential effects of halometasone on the therapeutic efifcacy of topical vitamin D3 analog in psoriasis.Methods HaCaT cells were treated by halometasone and calcipotriol with different concentrations for various durations. The cell proliferation was observed by CCK8 assay. Quantitative real-time PCR and Western blotting were used to detect the expression level of VDR mRNA and protein.ResultsHalometasone signiifcantly enhanced the inhibitory effects of calcipotriol on keratinocyte proliferation. Halometasone upregulated the expression of VDR at the level of mRNA and protein in a concentration-dependent manner.ConclusionThe upregulation of VDR expression by halometasone in keratinocytes may partly explain the phenomenon that halometasone could enhance the therapeutic efifcacy of topical vitamin D3 analog in the treatment of psoriasis.%目的:观察卤米松对角质形成细胞维生素D受体(vitamin D receptor,VDR)表达的影响,以探讨糖皮质激素对维生素D3衍生物治疗银屑病效应的潜在影响。方法采用CCK8法观察不同浓度卤米松、卡泊三醇在不同时相点下对HaCaT角质形成细胞增殖的影响;采用反转录-实时荧光定量聚合酶链反应(PCR)、蛋白印迹法,观察不同浓度卤米松在不同时相点作用下HaCaT细胞VDR的表达情况。结果卤米松可增强卡泊三醇对HaCaT细胞增殖的抑制作用;卤米松作用24 h、48 h后,HaCaT细胞中VDR mRNA及蛋白水平均明显升高,并呈剂量依赖性效应。结论卤米松上调VDR的表达可能有助于提高角质形成细胞对维生素D3衍生物作用的反应性,从而有可能提高维生素D3衍生物对角质形成细胞的治疗效应。
