粘着斑激酶基因沉默促进口腔癌细胞侵袭和迁移的实验研究

         

摘要

Objective:To invesligale the effecls of focal adhesion kinase (FAK) on the invasion and migration of human oral squa-mous cell carcinoma Tca8113 cells. Methods, RNA interference! RNAi) melhod with pSilencer-FAK veclor was used to inhibile FAK expression. The velocity of cell migration was measured by wound-repair experiment. The invasion of the cells was detected by invasion chamber assay. Results, FAK expression of the RNAi treated cells was suppressed by pSilencer-FAK vector. After FAK gene silencing, the velocity of migration and invasion of the cells were inhibited significantly. Conclusion, FAK can promote migration and invasion of Tca8113 cells.%目的:研究粘着斑激酶(focal adhesion kinase,FAK)对舌鳞癌细胞系Tca8113细胞侵袭和迁移的影响.方法:用RNA干扰的方法抑制Tca8113细胞FAK基因的表达.然后以划痕试验测定细胞迁移,以Transwell小室侵袭模型研究细胞侵袭能力.结果:外源性干扰质粒可以显著抑制Tca8113中FAK的表达;FAK基因沉默后,Tca8113细胞迁移能力、侵袭能力明显下降.结论:FAK在舌鳞癌Tca8113细胞的迁移和侵袭过程中发挥重要作用.
