PRM1基因C321A、G197T单核苷酸多态性对体外受精结局的影响

         

摘要

Objective:To explore the association between single nucleotide polymorphism (SNP) loci C321A & G197T in protamine 1 (PRM1) gene and IVF fertilization rate.Methods:A total of 182 IVF-ET cycles in our reproductive center were collected.The cycles were divided into three groups by the fertilization rate:the cycles with fertilization rate<30% in group A (n=43),30% to 65% in group B (n=24) cycles,and>65% in group C (n=115).The genomic DNA of residual sperm after insemination was extracted,and the frequency of C321A and G197T in PRM1 was detected by PCR-RFLP analysis.The differences in genotype and IVF fertilization rate were compared among the three groups.Results:There was no any significant difference in the genotype frequency of C321 polymorphism in PRM1 gene among the three groups(P>0.05).C321A polymorphism in PRM1 gene was not associated with IVF fertilization rate(P>0.05).The frequency of genotype GG,GT and TT of G197T in PRM1 gene was 9.3%(4),25.6%(11),and 65.1%(28)in group A;the frequency of genotype GG,GT and TT was 8.3%(2),25.0%(6),and 66.7%(16)in group B;and the frequency of genotype GG,GT and TT was 38.3%(44),26.1%(30),and 35.6%(41)in group C respectively.The frequency of genotype TT of G197T in group A and group B was significantly higher than that of group C(65.1% vs.35.6%,66.7% vs.35.6%)(P<0.05).Genotype TT of PRM1 G197T increased the risk of fertilization failure.Conclusions:The SNP of C321A in PRM1 is not associated with IVF fertilization rate.The SNP of G197T in PRM1 is associated with IVF fertilization rate.%目的 探讨鱼精蛋白1的C321A、G197T基因多态性与体外受精结局的关系.方法 选取我院生殖中心进行常规IVF的182例(182个周期),根据受精率分为3组:A组受精率<30%,43个周期;B组受精率在30%~65%之间,24个周期;C组受精率>65%,115个周期.提取授精后剩余精子的基因组DNA,采用聚合酶链限制性片段长度多态性(PCR-RFLP)方法检测PRM1基因C321A和G197T 基因型频率和分布,比较3组间基因型的分布差异及所有研究对象不同基因型间受精率的差异.结果 3组间比较,PRM1基因C321A多态性位点的各基因型频率无显著性差异(P>0.05),C321A多态性位点与IVF受精率无相关性(P>0.05).在A组中,PRM1基因 G197T多态性位点GG、GT、TT基因型分布频率分别为9.3%(4)、25.6%(11)、65.1%(28);B组分别为8.3%(2)、25.0%(6)、66.7%(16);C组分别为38.3%(44)、26.1%(30)、35.6%(41).A组和B组的TT基因型频率显著高于C组(65.1% vs.35.6%,66.7% vs.35.6%),差异均有统计学意义(P<0.05),TT基因型增加了IVF受精失败的风险.结论 PRM1基因 C321A多态性与IVF受精率不相关;PRM1基因 G197T多态性改变与IVF受精率相关.
