Objective:To study the effect of mifepristone on expression of BRCA1 and BRCA2 gene in breast cancer cells.Methods:The fluorescent real-time quantitative PCR method was used to detect the expression of BRCA1 and BRCA2 gene in the cells treated with mifepristone or not.Results:With the dose of mifepristone increased(0.25,2.5,25 μmol/L),both of the expressions of BRCA1 and BRCA2 mRNA in T47D cells were reduced.The expression of BRCA1 mRNA in the 25 μm group was significantly lower than that in the control group or the 0.25 μm group (P < 0.05).The expression of BRCA2 mRNA in the 0.25 μm group was significantly lower than that in the control group (P<0.05).BRCA2 mRNA was also significantly lower in the 25 μm group compared with the control group or the 0.25 μm group(P<0.01).Conclusions:Mifepristone inhibits the expressions of BRCA1 and BRCA2 at mRNA level in T47D cells in a dose-dependent manner.%目的 研究米非司酮处理乳腺癌细胞系T47D后,BRCA1和BRCA 2在mRNA水平的表达变化. 方法 使用不同浓度米非司酮(0.25、2.5、25 μmol/L)分别处理人乳腺癌细胞系T47D 1d后,应用荧光实时定量聚合酶链反应(Realtime-PCR)的方法检测各浓度组和对照组BRCA1、2基因的表达差异. 结果 不同浓度米非司酮(0.25、2.5、25 μmol/L)分别处理人乳腺癌细胞系T47D 1 d后,BRCA1、2基因在mRNA水平上表达均降低.米非司酮25 μmol/L组分别与对照组和0.25μmol/L组比较,BRCA1基因在mRNA水平上表达均降低,且具有统计学差异(P<0.05).与对照组比较,米非司酮2.5 μmol/L组BRCA2基因在mRNA水平上表达降低,且具有统计学差异(P<0.05);米非司酮25 μmol/L组分别与对照组和0.25 μmol/L组比较,BRCA2基因在mRNA水平上表达降低,均具有统计学差异(P<0.01). 结论 在乳腺癌细胞中,米非司酮在转录水平上调控BRCA1、2基因的表达.
