壳聚糖/膨润土固定化脂肪酶水解橄榄油的研究

         

摘要

In order to solve the low catalytic stability, difficult recovery and other issues of free lipase, we tried to choose the different ratio of chitosan/bentonite as the carrier of lipase to improve its activity in the hydrolysis of olive oil. The chitosan/bentonite composites were characterized by Fourier Transform Infrared Spectroscopy ( FT-IR) and Scanning Electron Microscopy ( SEM) , and the catalytic properties of immobilization lipases on various chitosan/bentonite in the hydrolysis of olive oil were studied. The results show that 10% of the mass ratio of chi-tosan and bentonite (CB-10) can achieve the best immobilization yield of lipase (92. 1%) on the carrier, and the catalytic activity of the immobilized enzyme is 0. 56U/mg. The residual activity of the immobilized enzyme is 68. 2% after 5 consecutive batches of the reaction, showing good catalytic stability. The research findings indi-cate that the chitosan/bentonite composite which can make a better balance between the structural rigidity of the inorganic material and the biocompatibility of the natural polymer material is a good carrier of the immobilized en-zyme.%为了改善游离脂肪酶催化稳定性低、难以回收等缺陷,以不同比例的壳聚糖/膨润土复合载体负载脂肪酶,以提高脂肪酶在橄榄油水解中的催化性能.通过红外光谱( FTIR)、扫描电镜( SEM)等对壳聚糖/膨润土复合材料进行表征,并考察了各种壳聚糖/膨润土固定化脂肪酶对橄榄油水解反应的催化性能.结果表明:当壳聚糖与膨润土的质量比为10%( CB-10)时,脂肪酶的负载量达到最高(92.1%),固定化酶的催化活性为0.56 U/mg,连续反应5批次后,固定化酶的剩余活性为初始值的68.2%,显示了较好的催化稳定性.该研究结果说明,壳聚糖/膨润土复合材料较好地兼顾了无机材料的结构刚性和天然高分子物质的生物相容性,是固定化酶的良好载体.
