环磷酰胺对糖尿病肾病大鼠肾组织中MCP-1、TGF-β1表达的影响

         

摘要

目的 探讨环磷酰胺(CTX)对2型糖尿病肾病大鼠肾组织中单核细胞趋化因子蛋白-1(MCP-1)及转化生长因子β1(TGF-β1)表达的影响,寻找治疗糖尿病肾病的新方法.方法 80只SD雄性大鼠随机分为对照组(N组)、高脂饮食组(HF组).HF组给予高脂饲料喂养,8周产生胰岛素抵抗后,一次性腹腔注射STZ(35 mg/kg),N组给予常规饲料喂养,腹腔注射等量的柠檬酸-柠檬酸钠缓冲液.2型糖尿病肾病造模成功后,将成模大鼠随机分为糖尿病肾病模型组(DN组)及治疗组(CTX组).CTX组腹腔注射CTX 30 mg/kg,N组及DN组给予等量的生理盐水腹腔注射.3组均为每周1次,连续4周.肾组织切片免疫组织化学染色检测肾组织中MCP-1、TGF-β1的表达,用彩色图像处理软件处理图像,计算平均吸光度.结果 ①DN组与N组比较:肾组织中的MCP-1、TGF-β1表达明显增强;②CTX治疗后糖尿病肾病大鼠肾组织中MCP-1及TGF-β1表达明显减轻,与DN组比较,P<0.05.结论 ①MCP-1、TGF-β1可能参与了糖尿病肾病的发生;②CTX能够减轻糖尿病肾病肾组织中MCP-1、TGF-β1的表达.%Objective To investigate effects of cyclophosphamide (CTX) on the expressions of monocyte chemokine protein-1 (MCP-1) and transforming growth factor betal (TGF-β1 ) in the renal tissues of type 2 diabetic nephropathy and to find a new approach to diabetic nephropathy treatment. Methods Totally 80 SD rats were randomly divided into control group (N group) and high-fat diet group (HF group). HF group was given a single intraperitoneal injection of STZ (35 mg/kg) afer insulin resistance in week 8 through high-fat diet daily. At the same time, N group was given conventional feed and intraperitoneal injection of the same amount of citric acid-sodium citrate buffer. The film-forming rats were then randomly divided into diabetic nephropathy model group (DN group) and treatment group (CTX group) after type 2 diabetic nephropathy model was successfully established. CTX group was given intraperitoneal injection of CTX (30 mg/kg) while N group and DN group were given the same amount of saline by intraperitoneal injection. Administration in the three groups was done once a week for four consecutive weeks. MCP-1 and TGF-β1 expressions in the renal tissue were detected by immunohistochmical staining, images were processed with color image processing software to calculate the average optical density. Results ① The expressions of MCP-1 and TGF-β1 in DN group were significantly enhanced compared with those in N group. ② The expressions of MCP-1 and TGF-β1 in diabetic nephropathy after CTX treatment were significantly reduced compared with those in DN group (P<0.05). Conclusion ① MCP-1 and TGF-β1 may be involved in the occurrence of diabetic nephropathy. ② CTX can reduce the expressions of TGF-β1 and MCP-1 in the diabetic renal tissues.
