断血流皂苷A提取工艺研究

         

摘要

目的:通过设计正交试验,研究断血流皂苷A的最优提取工艺.方法:采用C18色谱柱(150 mm×4.6mm,5μm),流动相为甲醇体积:水体积(80:20),柱温为30℃,流速1.00 mL/min,紫外检测器波长为250nm.考察溶剂浓度、提取时间、溶剂用量对血流皂苷A提取的影响.实验结果:采用75%乙醇为提取溶剂,加入200倍溶剂,超声提取30 min为最优提取方法.结论:该方法合理可行,可用于断血流的质量控制.%Orthogonal experimental design is used to find out the optimal extraction conditions for Clinoposide A in this paper. A C18 column (ISO mmx4. 6 mm, 5 u,m)is employed as high performance liquid chromatography (HPLC). The mobile phase is methanol/ water with flow rate of 1.00 mL/min, column temperature of 30 t! and the wavelength of ultralvolet detector is 250 nm. The concentrations of ethanol, extraction time, the amount of ethanol are considered as the main factors affecting the result. The experimental results show that the optimal condition is that 75% ethanol is used with 200 times solvent and coped by ultrasound for 30 min. The method is reasonable and feasible, it can be used in the quality control of clinoposide concrete.
