异甘草素对人宫颈癌细胞增殖和凋亡的影响

         

摘要

目的:研究甘草查尔酮类活性成分异甘草素(Isoliquiritigenin,ISL)对人宫颈癌 Siha 细胞增殖和凋亡的影响。方法将细胞接种于96孔板进行培养(每个孔的密度为1×104个/mL),待细胞完全贴壁后(24 h),将细胞分为 ISL 25、50、100、200、300、400μg/mL 组及对照组(正常培养的 Siha 细胞)、顺铂组(顺铂25μg/mL)、DMSO 组(20μL DMSO 加入5 mL 的培养基中),用 MTT 法测定 ISL 对人宫颈癌 Siha 细胞增殖的抑制作用。通过流式细胞仪测定细胞凋亡及细胞线粒体跨膜电位的变化。通过 Caspase-3、Caspase-8、Caspase-9活性检测试剂盒测定 ISL 对半胱天冬酶活性的影响。结果ISL 抑制 Siha 细胞增殖呈时间依赖性及浓度依赖性,可浓度依赖性的升高细胞凋亡率,降低线粒体膜电位,能增强 Caspase-3的表达活性,但对 Caspase-8、Caspase-9的活性不产生影响。结论ISL 能抑制宫颈癌细胞的增殖,其可能机制是诱导细胞线粒体膜电位途径实现的。%Objective To study the effect of licorice active component Isoliquiritigenin (ISL)on human cer-vical cancer SiHa cell proliferation and its apoptosis.Methods Cells were seeded in 96 well plates and cul-tured (destiny of each well was 1×104 ),after the cells were fully adherent (24 h),the cells were divided into 4 groups,ISL group:ISL 25,50,100,200,300,400 μg/mL,control group is normal cultured cells of siha,positive control group (Cisplatin 25 μg/mL),negitive control group (20 μL DMSO was added 5 mL medium).MTT assay was used to examine the inhibitory effect of ILG towards cervical cancer SiHa cell growth.The flow cytometry (FCM)was used to examine the ILS effect to induction of the cancer cell apoptosis and the effect to mitochondrial membrane potential (MMP).The impact of ILG to caspases was measured by caspase-3,caspase-8,caspase-9 activity assay kits.Results ISL significantly inhibited the growth of SiHa cells in a concentration-dependent manner.ISL increased apoptosics and decreased mito-chondrial membrane potential.The induction of apoptosis in human cervical cancer SiHa cell whether by activating caspase enzymes apoptotic pathway still needs to further study.Conclusion These results sug-gest that the mitochondrial aerobic respiration and Caspase-3,8,9 activation shift could contribute to the action of ISL against Siha cancer cells.
