垂直胸部撞击致兔急性肺损伤动物模型的建立

         

摘要

Objective To establish the animal model of rabbit acute lung injury(ALI)of vertical chest impact trauma and to provide the reference to the related experimental study.Methods 6 7 New Zealand white rabbits were randomly divided into four groups,with mild shock,moderate shock group and the se-vere impact and control groups.The rabbits were hit by obj ects with different weight and ground clearance in the way of free fall,in order to make vertical impact on the right chest,causing acute lung inj ury.Then the vertical impact index was calculated.Results When the vertical impact index was from 30 to 60,the impact of the 4 h after oxygenation index(238 ± 29)was significantly different (P <0.05)from the pre-impact oxygenation index(422 ± 15),thus it was feasible to establish a stable animal model of acute lung injury in rabbits.When vertical impact index> 60,the number of deaths in severe impact(nine cases)was significantly different (P<0.05).from the number(four cases)in moderate impact group.Conclusion U-sing vertical impact to establish animal model of acute lung inj ury in rabbits is with typical pathological changes,easy operating,needs no special laboratory equipment,which is conducive to teaching and re-search work of grass-roots hospitals for acute lung inj ury,and provides experimental and theoretical basis for the occurrence and development of chest impact trauma of acute lung inj ury(ALI).%目的建立垂直胸部撞击致兔急性肺损伤动物模型,为胸部撞击致急性肺损伤的实验研究提供参考依据。方法随机选取新西兰大白兔67只,分为轻度撞击组、中度撞击组、重度撞击组和对照组4组,用不同质量和离地高度的撞击物,以自由落体的方式,垂直撞击右侧胸部,造成急性肺损伤,并计算垂直撞击指数。结果当垂直撞击指数为30~60时,撞击后4 h的氧合指数(238±29)与撞击前氧合指数(422±15)比较差异有统计学意义(P <0.05),可建立稳定的兔急性肺损伤动物模型。当垂直撞击指数>60时,重度撞击组实验兔的死亡率与中度撞击组比较差异有统计学意义(P <0.05)。结论采用垂直胸部撞击方式建立的兔急性肺损伤动物模型,病变典型,操作简便,无需特殊实验设备,有利于基层医院对急性肺损伤的教学和科研工作,为胸部撞击致急性肺损伤的发生、发展提供实验和理论依据。
