杏鲍菇结构物质预防脂肪肝和降血糖的研究

         

摘要

设计不同剂量的杏鲍菇纤维素分别联合高脂饲料饲养大鼠,以考来烯胺为阳性对照,研究杏鲍菇纤维素对大鼠高脂血症性脂肪肝预防作用,预防5周后检测大鼠血清中ALT、AST、TC、TG水平,肝脏组织中MDA、SOD活性及观察肝组织病理学变化情况.实验结果:杏鲍菇纤维素能降低大鼠血清中ALT、AST、TC、TG水平(P<0.01),显著提高大鼠肝脏组织中SOD活性、降低MDA含量(P<0.05),且具有剂量效应.肉眼观察模型组肝脏体积明显增大,整个肝脏呈奶黄色,而喂养杏鲍菇纤维素的肝脏有不同程度的好转;组织病理学也发现杏鲍菇纤维素各组的肝细胞无脂肪变性,无脂肪空泡,并值得注意是,与考来烯胺组相比较,杏鲍菇纤维素高剂量组的预防效果与之相近,无差异性(P>0.05),实验结果显示,杏鲍菇纤维素具有预防高脂血症性脂肪肝形成的作用;采用打击高血糖模型检测杏鲍菇浸提物的体内降血糖效果,结果发现:与模型对照组比较,各剂量组大鼠血糖的下降百分率显著性增高,其中高剂量组达到极显著性(P<0.001);在糖耐量试验中,高、中剂量组大鼠糖耐量与模型对照组比较均呈较显著差异(P<0.01),低剂量组大鼠糖耐量与模型对照组比较呈显著差异(P<0.05);高剂量对正常大鼠空腹血糖无明显影响(P>0.05).实验结果显示,杏鲍菇浸提物具有辅助降血糖功能.%To investigate the preventive effect of Pleurotus eryngii cellulose on fatty liver rats with hyperlipemia, the rats were fed with different dose of the cellulose coupled with high fat diet with cholestyramine as positive control. After 5 weeks, the levels of ALT, AST, TC and TG in serum, and the activities of malonaldehyde (MDA), and superoxide dismutase(SOD) were detected; and the pathological changes in the liver were observed. The results showed that P. eryngii cellulose could significantly lower the ALT, AST, TC and TG concentration in serum (P<0.01), and improved the activity of SOD while significantly decreased the content of MDA (P<0.05) in a dose dependent manner. In addition, the histopathological test showed that the livers of the negative group (treated without cellulose) were milk-yellow and bigger, and there was no fatty generation or vacuoles in the livers of the administration groups. Notably, the preventive effect of the cellulose is similar to that of cholestyramine (P<0.05). All these results indicated that the cellulose of Pleurotus eryngii has the potential of preventing the fatty liver with hyperlipemia in rats. The hypoglycemic effect of P. eryngii extracts was carried out on the model of the hyperglycemia mice in vivo. The decline percentage of blood glucose in each model dose group had significant differences than that of model control group (P<0.05),and the high dose group reached a very significant(P<0.001). Compared with the model control group, the glucose tolerance in high dose group and medium dose group were all more significantly difference(P<0.01),and the glucose tolerance in low dose group was significantly difference(P<0.05). High dose normal group had no significant effect on the fasting blood glucose with normal control group (P>0.05). So P. eryngii extracts had the hypoglycemic function of hyperglycemic animal.
