响应面法优化水蜈蚣总黄酮的提取及其抗氧化性研究

         

摘要

[Objective] To determine and optimize the extraction technology of total flavonoids from Kyl inga Brevifolia Rottb and antioxidant activity. [Methods] The optimal extraction parameters and the extraction rate were studied by response surface methodology. Select the ethanol concentration, extraction temperature, ratio of material to liquid and extracting time as factors, and the extraction rate of total flavonoids as the response value, the experimental design of four factors and three levels was applied to Box-Behnken central composite method. [Results] The optimal conditions for the extraction rate of total flavonoids were 60% ethanol concentration, extraction temperature at 70 ℃, extraction time 5h, ratio of solid to liquid 1:40. and the flavonoids yield was up to 1.969% at optimal conditions. The relative error of the experimental values and regression model to predict the values was 1.68%, and the hydroxyl radical scavenging rate at 25.1%. [Conclusion] Optimization of extracting technology condition was reliable, which based on response surface method, and it had good scavenging effect on hydroxyl free radical.%[目的]确定水蜈蚣总黄酮乙醇提取的最佳工艺条件及其抗氧化性。[方法]采用响应面法优化乙醇提取水蜈蚣总黄酮的条件。在单因素实验基础上,选取乙醇浓度、提取温度、料液比和提取时间为影响因子,应用Box-Behnken中心组合法进行四因素三水平的试验设计,以水蜈蚣黄酮得率为响应值,进行响应面分析。[结果]乙醇提取水蜈蚣的最佳工艺条件为乙醇浓度60%,提取温度70℃,提取时间5 h,料液比1:40,在优化工艺下,水蜈蚣黄酮的得率达1.969%。实验值与回归模型预测值的相对误差为1.68%。水蜈蚣总黄酮对羟自由基的清除率达25.1%。[结论]基于响应面法分析所得的优化提取工艺条件可靠,水蜈蚣总黄酮对羟自由基的清除作用较好。
