线粒体在骨关节炎发展中的作用及机制

         

摘要

骨关节炎(OA)是最常见的年龄相关性关节病,其发病机制复杂.目前认为关节软骨发生的代谢和结构变化在OA的发生和进展中起主要作用.线粒体是真核细胞中最重要的细胞器之一,在诸多生理活动中起重要作用,与细胞凋亡、老化和一系列的病理过程密切相关.最近的研究表明,线粒体在OA发病机制中发挥作用.线粒体DNA单倍群是以线粒体DNA序列中特定单核苷酸多态性为特征的单个组群,是一种遗传变异,被认为参与OA的发生、发展; 线粒体功能障碍与凋亡、衰老以及多种病理状态有关,线粒体功能障碍影响软骨细胞的呼吸链活性,增加炎症反应,促进细胞凋亡,是导致OA发生的重要因素之一.%Osteoarthritis(OA) is the most common age-related joint disease and the pathogenesis of OA is complex. The metabolic and structural changes that take place in the articular cartilage are thought to play a main role in the initiation and progression of OA. The mitochondrion is one of the most important organelles found in eukaryotic cells,which is associated with apoptosis, aging, and a number of pathological conditions. Recent studies have indicated a role for mitochondria in the development of OA. Mitochondrial DNA haplogroup is a single group characterized by specific single nucleotide polymorphisms in mitochondrial DNA sequences,which is a genetic variation and is considered to be involved in the occurrence and development of osteoarthritis; mitochondrial dysfunction is associated with apoptosis, aging and various pathological conditions. Mitochondrial dysfunction affects the respiratory chain activity of chondrocytes,increases inflammatory response and promotes apoptosis,which is one of the important factors leading to osteoarthritis.
