Objective:To investigate the inhibitory effects of Pinus massoniana bark extract (PMBE) on tumor metastasis in breast cancer MCF-7 cells in vitro.Methods:Effects of PMBE on the viability of breast cancer MCF-7 cells were detected by MTT assay.Breast cancer cell metastasis after exposure to PMBE were quantified by wound healing and Transwell assays,respectively.The expression of MMM-9 protein were evaluated by western blot.Results:Breast cancer cells after PMBPs treatment showed a dramatic reduction in cell viability in a dose-dependent manner,the IC50 value of PMBP was (71±2.1) μg·mL-1.After cells were treated with 5 and 15 μg·mL-1 PMBE,the cells invasion potentials were also decreased by 25% and 36%,the cell metastatic potentials were also decreased by 59% and 79%,respectively.Conclusion:Our findings indicated that PMBE had potential to develop as an anti-metastatic cancer drug for breast cancer patients.%目的:研究松树皮提取物原花青素(PMBE)对人乳腺癌MCF-7细胞体外转移及侵袭作用的影响,并初步探讨其作用机制.方法:不同浓度PMBE作用于体外培养的人乳腺癌MCF-7细胞,采用MTT法检测其对细胞增殖抑制作用的影响;通过划痕实验检测PMBE对细胞转移能力的影响;体外侵袭实验(Transwell小室)检测PMBE对细胞侵袭能力的影响;最后应用免疫印迹(Western-blot)方法检测PMBE对MCF-7细胞基质金属蛋白酶-9表达水平的影响.结果:随着PMBE浓度的增加,MTT法检测结果表明,PMBE在体外对MCF-7细胞有明显的增殖抑制作用,半数致死浓度(IC50)值为(71±2.1)μg·mL-1;PMBE质量浓度为5、15 μg·mL-1时,细胞划痕的闭合能力分别下降25%、36%,细胞侵袭能力下降59%、79%;PMBE能显著抑制MCF-7细胞MMP-9蛋白的表达.结论:PMBE能有效抑制乳腺癌增殖和转移,其作用机制可能与降低乳腺癌细胞运动能力有关,PMBE有潜力开发为治疗乳腺癌的药物.
