Objective:To study effect of hesperidin on product of extracellular matrix in human embryonic lung fibroblast stimulated by TGF-β1.Methods:12.5-200 μmol ·L-1 hesperidin was cultured with MRC-5 cells,a sort of human embryonic lung fibroblast,aiming at finding dose extent of hesperidin without significant effect on cellular viability.MRC-5 cells were stimulated with 5 ng· mL-1 TGF-β1 for increase production of extracellular matrix,at the same time,hesperidin was used with TGF-β1.Results:12.5-50 μmol ·L-1 hesperidin was not significant effect on viability of MRC-5 cells(P >0.05).The dose of hesperidin was significantly decreased o-SMA,Col-I and FN levels (P < 0.05).Conclusion:Hesperidin could inhibit production of extra cellular matrix in MRC-5 cells stimulated by TGF-β1.%目的:探讨橙皮苷对TGF-β1诱导的人胚肺成纤维细胞合成细胞外基质的影响.方法:12.5~200μmol·L-1的橙皮苷作用于MRC-5细胞,得出不影响细胞增殖的剂量用于后续实验.5 ng·mL-1的TGF-β1作用于MRC-5细胞,诱导细胞大量合成或分泌细胞外基质成分,同时用橙皮苷干预.结果:12.5~50 μmol·L-1橙皮苷未发现显著影响MRC-5细胞的增殖(P>0.05).受到TGF-β1刺激后,MRC-5细胞的Col-I分泌量和α-SMA及FN的合成量均显著增加(P<0.05).12.5~50μmol·L-1橙皮苷显著减少上述分子的分泌量或合成量(P<0.05).结论:橙皮苷抑制TGF-β1诱导的人胚肺成纤维细胞合成或分泌细胞外基质成分.
