Objective To observe the expression and significance of PTEN and PDCD4 in gastric cancer. Methods Immuno⁃histochemical SP method was used to determine the expression of PTEN and PDCD4 in gastric cancer and paired adjacent normal mucosa tissues. The relationships between PTEN, PDCD4 and clinicopathological characteristics, prognosis of gastric cancer were analyzed. Results Thepositive expression rate of PTEN and PDCD4 in gastric cancer was significantly lower in normal gastric tissues ( P <0. 05). The expression of PTEN was closely correlated with tissue differentiation,tumor depth,lymph node metastasis and TNM stages of the cancer (P<0. 05). The expression of PDCD4 was closely correlated with tissue differentiation,tumor depth,lymph node metastasis and TNM stages of the cancer (P<0. 05). PTEN had positive correlation with PDCD4 in gastric cancer (r=0. 801,P<0. 05). The total five-year survival rate was 45. 3% in 75 cases of patients. Univariate and multivariate analysis showed that lymph node metastasis was independent prognostic factor for patients with gastric cancer (P<0. 05). Conclusion The expression of PTEN and PDCD4 might be correlated to the cancer and development of gastric cancer. Lymph node metastasis is independent prognostic factor for patients with gas⁃tric cancer.%目的：探讨第10号染色体同源缺失性磷酸酶－张力蛋白基因（ PTEN）和程序性细胞死亡因子4（PDCD4）在胃癌组织中的表达及意义。方法应用免疫组化SP法，检测75例胃癌组织、35例相对应的癌旁正常组织中PTEN、PDCD4蛋白的表达情况，分析PTEN、PDCD4的表达与临床病理特征及预后的关系。结果 PTEN、PDCD4在胃癌组织中的表达均明显低于癌旁正常组织，差异有统计学意义（P＜0．05）。 PTEN阳性表达率与组织分化程度、TNM分期、浸润深度、淋巴结转移相关（P＜0．05），PDCD4的阳性表达率与组织分化程度、TNM分期、浸润深度、淋巴结转移相关（P＜0．05）；PTEN、PDCD4在胃癌组织中的表达呈正相关性（r＝0．801，P＜0．05）。75例患者5年总生存率为45．3％。单因素和多因素生存分析显示，淋巴结转移是胃癌患者预后的独立风险因素（ P＜0．05）。结论 PTEN、PDCD4的低表达与胃癌的发生、发展有关，淋巴结转移是胃癌患者预后的独立风险因素。
