目的：观察双醋瑞因对黄韧带肥厚的作用机制，探讨应用双醋瑞因预防和治疗黄韧带肥厚的效果。方法对黄韧带肥厚的大白兔分别应用双醋瑞因作药物组（45只），与生理盐水灌胃做模型组（45只），正常大白兔45只作为正常对照组；于第4、8和12周，随机处死各组动物至少15只，取黄韧带，采用免疫组化的方法观察三组免疫组化切片中IL－1、IL－6及TNF－α的表达情况。结果模型组与药物组中黄韧带细胞质或细胞核中IL－1、IL－6、TNF－α的表达明显高于正常组（P＜0．05）。药物组的IL－1、IL－6、TNF－α表达阳性率显著低于模型组（P＜0．05）。结论双醋瑞因对局部组织的炎症因子起到了改善作用；通过双醋瑞因的药物干预，肥厚黄韧带中胶原纤维增生明显受到抑制，从而改善了肥厚黄韧带中的组织结构，可使之恢复至正常状态或接近正常状态。%Objective To observe the action mechanism of diacerein on hypertrophy of ligamentum flavum,and explore prevention and treatment of diacerein for ligamentum flavum hypertrophy. Methods The flavum ligament hypertrophy of rabbits were respectively given diacerein (as drug group,n=45),normal saline viastomach(n=45,as model group),and 45 normal rabbits as normal control group;at the fourth,eighth and twelfth weeks,each group were randomly sacrificed animal at least 15,the flavum ligament was taken out,the three groups were detected in their IL -1,IL -6 and TNF -alpha in slice mining with the methods of immunohistochemistry. Results The expression of flavum ligament cytoplasm or nucleus IL-1,IL-6,TNF-alpha in model group and drug group was significantly higher than that in normal group,there was statistical difference(P<0.05). The positive expression of IL-1,IL-6,TNF-alpha rate in drug group was significantly lower than that in model group,there was statistical difference(P<0.05). Conclusion Diacerein plays a positive improvement on local tissue inflammation factor,by diacerin intervention,in ligamentum flavum hypertrophy collagen fiber hyperplasia is obviously inhibited,thereby improving the organization structure of the ligamentum flavum hypertrophy,so restored to normal state or close to normal state.
