氯胺酮预处理对卵蛋白激活的致敏大鼠肺泡巨噬细胞丙二醛及超氧化物歧化酶的影响

         

摘要

Objective To study the effects of different doses of ketamine on malondialdehyde (MDA) and superoxide dismutase (SOD) in alveolar macrophages of rats sensitized by ovalbumin (OVA).Methods Alveolar macrophages were isolated and cultured,and pretreated with ketamine at concentration of 10,100 and 1 000 μmol/L,then challenged with OVA.After being cultured for 72 h,the production of SOD and MDA in the supernatant of macrophage was assayed.Results Compared with control group,the MDA level in culture supernatants of OVA group was significantly increased with the decreased level of SOD,while higher concentration of ketamine (100 and 1 000 μmol/L) pretreatment could inhibit this response.Conclusion Ketamine pretreatment exerts a protective effects on OVA-induced oxidative stress injury of alveolar macrophage.%目的 研究不同剂量氯胺酮对卵蛋白(OVA)刺激致敏的大鼠肺泡巨噬细胞丙二醛(MDA)和超氧化物歧化酶(SOD)的影响.方法 致敏的大鼠肺泡巨噬细胞分离培养,氯胺酮(10、100、1 000 μmol/L)预处理后给予OVA刺激,培养72 h后检测细胞培养上清液中MDA及SOD水平.结果 与对照组比较,OVA组培养上清液中MDA水平明显增加,伴有SOD水平下降,而较高浓度氯胺酮预处理(100、1 000 μmol/L)后可起到一定的抑制作用.结论 氯胺酮预处理对OVA引起的肺泡巨噬细胞氧化应激损伤具有一定的保护作用.
