原位裸小鼠肝癌模型的建立及高场MR成像的实验研究

         

摘要

目的:直接注射法制作裸小鼠肝癌模型,小动物专用高场MR(7.0T)观察其表现.方法:抽取H22肝癌荷瘤鼠腹水,直接注射法种植于20只裸小鼠肝脏,术前及术后不同时间应用MR对种植肿瘤进行观察,并与病理切片对照分析.结果:20只模型小鼠肝脏均可见肿瘤病灶,移植成功率100%,3d后MR检查可见单发肿瘤病灶,直径约(0.6±0.2) mm,病理特征与人类肝癌相似,均为肝细胞性肝癌.结论:直接注射法制作裸小鼠肝癌模型简便、易行、成功率高,高场MR可对移植瘤进行动态观察,为肿瘤的早期诊断及药物疗效评价提供实验基础.%Objective:To establish implanted liver tumor in nude mice with immediate injection method and to observe the process of tumor formation with highfield MR scanner used specially for small animals. Methods:Cancerous ascites of H22 cell mice was injected into the liver of 20 nude mice directly. The formation of tumor was observed by MR and MRI. And the findings were checked with pathological examinations. Results; The formation ratio of tumor with immediate injection method was high (100%). MR imaging showed that single tumor was seen in the liver of all mice three days later. The average diameter of nodes was (0. 6±0. 2)mm. The pathologic characteristics of liver tumor were similar to those of the human's and all the tumors were proved to be hepatocellular carcinoma. Conclusion:Direct injection is a simple method for establishing liver tumor model and the formation process of tumor can be observed by highfield MR clearly. It will provide experiment foundation for assessment of early diagnosis and therapy of liver tumor.
