The level of free proline content in plant tissues is often regarded as a physiological index indicating the strength of plant anti-adversity. This paper explores and analyzes the reasons and elimination methods of the extraction and emulsification phe-nomenon of toluene in the process of determining the index by acidic ninhydrin colorimetry method, hoping to provide refer-ences for scientific researchers and students engaging in the de-termination of the index.%植物组织中游离脯氨酸含量的高低常常作为反映植物抗逆性强弱的一项生理指标，本文探讨分析了酸性茚三酮显色法测定该指标过程中甲苯萃取乳化现象产生的原因及其消除方法，希望能给从事该指标测定的科研工作者和学生提供参考。
