红景天苷改善血管内皮舒缩功能实验研究

         

摘要

目的 :研究低氧环境下红景天苷对血管内皮细胞舒缩功能的影响及其机制.方法:空白对照组内皮细胞暴露正常氧环境,低氧组、低氧和红景天苷干预组血管内皮细胞暴露于低氧环境,检测血管内皮细胞HIF-1α、内皮素-1、血管内皮NO合成酶基因的表达变化.同时检测采用siRNA干扰HIF-1α后内皮舒缩因子表达变化.结果:与空白对照组比较,低氧组血管内皮细胞HIF-1α、内皮素-1表达显著增加(P<0.05),而血管内皮NO合成酶表达下降(P<0.05);与低氧组比较,低氧及红景天苷干预组血管内皮细胞内皮细胞HIF-1α、内皮素-1表达受抑制(P<0.05),而血管内皮NO合成酶表达增加(P<0.05).siRNA干扰HIF-1α显示,红景天苷通过调控HIF-1α调节内皮细胞舒缩功能(P<0.05).结论:红景天苷通过抑制HIF-1α基因表达,抑制血管内皮收缩因子表达及促进血管内皮舒张因子表达,从而改善血管内皮舒缩功能.%Objective :To evaluate the effect of the hypoxia environment and salidroside on the function of vas-cular endothelial cells .Methods:The blank control group exposed to the normal oxygen environment ,hypoxia group ,hypoxia and the intervention group vascular endothelial cells exposed to hypoxia environment ,detection the gene expression changes of HIF-1α,endothelin-1 ,vascular endothelial NO synthase in vascular endothelial cells . The expression of HIF-1α,endothelin-1 ,vascular endothelial NO synthase was detected by siRNA interference .Re-sults :Compared with the blank control group ,the expression of HIF-1α and endothelin -1 were significantly in-creased in vascular endothelial cells in hypoxia group ,while vascular endothelial NO synthase expression was de-creased .SiRNA interference HIF-1αexperiments showed that salidroside improve the function of endothelial cell by regulating HIF-1α.Conclusion:These findings imply that salidroside improvs the function of endothelial cell by reg-ulating ,through regulation the expression of ET-1 ,eNOS via a HIF-1α-dependent pathway .
