目的：探讨健康直系亲体负载肿瘤干细胞抗原的靶向DC‐CIK细胞的制备方法及对恶性肿瘤患者的治疗价值。方法：43例恶性肿瘤患者，用智能单采机采集健康直系亲体的外周血单个核细胞，其中贴壁细胞经体外诱导产生树突状细胞（DC ），悬浮细胞经体外诱导产生CIK ，DC负载不同肿瘤干细胞抗原制备靶向DC 细胞，将靶向DC与CIK细胞共培养制备靶向DC‐CIK ，检测靶向DC‐CIK细胞表面分子的表达及细胞增殖情况，并将靶向DC‐CIK分6次回输给43例患者，治疗3个疗程后，结合影像学等检查综合评价临床治疗的疗效，同时观察不良反应。结果：靶向DC细胞与CIK细胞共培养后，可明显刺激CIK 细胞的增殖，同时CD3＋ CD8＋细胞，CD56＋及CD3＋CD56＋细胞的含量明显增加，43例恶性肿瘤患者接受健康直系亲体靶向DC‐CIK细胞输注，PR17例，SD 25例，PD 1例，临床疗效明显，无不良反应。结论：健康直系亲体靶向DC‐CIK细胞应用于恶性肿瘤患者效果确切、无明显不良反应。%Objective:To investigate the clinical efficacy of dendritic cells( DCs) sensitization by antigen from cancer stem cells in combination with cytokine‐induced killer (CIK) cells on patients with advanced cancer and detect their immune functions and adverse effects .Methods:The study analyzed retrospectively 43 patients with ad‐vanced cancer admitted in our department .Peripheral blood mononuclear cells (PBMCs) were isolated from Immedi‐ate children health of the above patients .Adherent cells were cultured to produce DCs .The DCs were pulsed with an‐tigens from cancer stem cells to produce target DCs .T lymphocytes were cultured to prepare CIK .The target DCs was co‐cultured with CIK to produce target DC‐CIK ,and then the phenotypes of target DC‐CIK were evaluated .The target DC‐CIK cell proliferation was determine ,too .The 43 patients received the immunotherapy of target DC‐CIK for 3 circles .After treatment ,the changes of T cell subtypes and cytokines released in the peripheral blood were eval‐uated .The therapeutic outcomes were evaluated .The adverse effects were also observed .Results:CIK cell prolifera‐tion was significantly enhanced after co‐cultured with target DCs .Meanwhile ,the percentages of CD3+ CD8+ cells , CD56+ and CD3+CD56+ cells went up significantly .Clinical responses of the target DC‐CIK treatment group were good .No severe adverse effects were seen in the patients .Conclusion:DCs derived from PBMCs of Immediate chil‐dren health of advanced cancer patients harboring antigens from cancer stem cells can differentiate into mature DCs , which facilitate the proliferation of CIK cells .The targrt DC‐CIK therapy improves the immune status of patients with advanced cancer .
