Objective :To evaluate the dynamic changes in expression of Toll like receptor on cornea epi‐thelial cells induced by lipopolysaccharide and illustrate the effect of TLR in the bacterial infection and Immune ho‐meostasis .Methods :Immortalized human corneal epithelial cell was treated by 10ng/ml and 1μg/ml lipopolysaccha‐ride .The expression of TLR2 and 4 were detected with qRT‐PCR from mRNA level and FACS from protein level , at 2 ,4 ,and 8 hours after stimulation .The IL‐6 level was examined by ELISA at 4 ,8 ,and 12 hours after stimula‐tion.Results :After stimulation with LPS ,cells elicited an increase in the content of TLR2 and 4 specific mRNA at each time point .The cell surface expression of TLR and IL‐6 secretion increased at early stage but decreased at late stage of LPS exposure .Conclusion :TLR 2 and 4 cell surface expression in HCE cell can recognize the stimulation of LPS ,and their expression pattern constitutes a negative‐feedback loop that is important in the bacterial infection and Immune homeostasis environment in the human cornea .%目的:观察角膜上皮细胞在细菌脂多糖作用后不同时间 Toll样受体(TLR)2、4表达的动态变化,初步明确T LR在角膜细菌感染和保持免疫稳态中的作用。方法:培养永生化人角膜上皮细胞系,分别用10ng/ml和1μg/ml的细菌脂多糖(LPS)刺激,作用2h、4h、8h后检测 TLR2、4的表达,4h、8h、12h后检测细胞分泌IL‐6能力。结果:不同浓度LPS 刺激后,细胞 TLR2、4的mRNA表达水平在不同时点均明显增高,但随着刺激时间延长,细胞表面蛋白表达和IL‐6的分泌水平则呈现先升后降的趋势。结论:人角膜上皮细胞(HCE)能够表达功能性的 TLR2、4,并且在接受刺激后,TLR蛋白表达和炎症因子分泌具有负反馈抑制。
