线粒体通路对骨关节运动损伤发生机制及预防措施研究

         

摘要

目的:研究膝关节软骨发生线粒体凋亡,其通路中各种相关因子对蛋白的表达,并对大鼠出现运动性的骨关节损伤同 HWTX-I相关的分子机制进行分析。方法:于实验大鼠的右膝关节位置切断前后交叉韧带,并结合进行中等强度的运动训练,来进行大鼠骨关节出现慢性损伤类模型的制作,同时与实施硬脊外腔进行置管术联合,进行重复性稳定的实验大鼠出现骨关节损伤为运动所致模型的构建。并基于此模型的基础上同布洛芬组作阳性对照,硬膜外腔应用 HWTX-I ,观察建模14d(早期)与建模28d(晚期)大鼠骨关节出现摩损性运动病变时,机体血清当中的谷胱甘肽过氧化物酶(GSH-PX),以及血清当中的超氧化物歧化酶(SOD)浓度出现的改变。结果:在硬膜外腔予以HWTX-I ,不仅能够形成抗伤害的显著效果,能够缓解及抑制骨关节慢性运动型损伤的病程进展,并且效果优于布洛芬,呈显著差异(P<0.01)。结论:硬脊膜外腔应用 HWTX-I ,能够使血清中的GSH-PX与SOD活力上调,对自由基产生的影响显著减轻。线粒体通路同骨关节的运动性损伤具有密切的相关性,能够指导防治骨关节损伤的进一步研究。%Objective :To study the articular cartilage of the knee occurred mitochondrial apoptosis ,ex-pression of the pathway related factors on the protein ,the blood free radicals may appear what kind of change ,and with reference to the signal pathway of mitochondrial apoptosis in the body of rats ,exercise bone joint injury and molecular mechanism of HWTX-I related analysis .Methods:The right knee position in experimental rats anterior and posterior cruciate ligament ,and combining the training of moderate intensity ,to make chronic injury rat model of bone and joint appeared ,at the same time combined with catheter and implementation of spinal cavity of rats ,the stability of repetitive construction of bone and joint injury appeared as the exercise model .And based on the basis of this model with ibuprofen group as positive control ,epidural HWTX-I modeling ,observe 14 days (early) and mod-eling for 28 days (late) rats bone joint friction movement lesions ,serum ,GSH-PX ,and the change of SOD concen-tration of the serum .Results :In the epidural space to HWTX-I ,not only can form a significant effects of anti inju-ry ,could progress alleviate and course of suppression of chronic exercise of bone and joint injury ,and the effect is better than that of ibuprofen , showed significant difference ( P< 0 .01 ) . Conclusion:Epidural application of HWTX-I ,can make the upregulation of GSH-PX and SOD activity in serum ,the impact on the free radicals signifi-cantly reduce .Has a close relationship with the movement of bone and joint injury of mitochondrial pathway ,can guide the further study on prevention and treatment of bone and joint injury .
