Objective: To discuss the effects of glycolysis inhibitor 2-Deoxyglucose combining with cisplatin against different types of pancreatic cancer cells, and to detect the preliminary mechanism about it. Methods:Cells (MiaPaCa2 & panc 1 ) incubated with lmg/L 2-Deoxyglucose and cisplatin. A apoptosis and Caspase 3-detected were done in 8 hours. After 24 and 48 hours, cells were under MTT detect. Results: Having positive inhibit effect on pancreatic cancer cells, 2-Deoxyglucose's anti cancer effect has an time-depend-effect, and combining with cisplatin, it rised its effect on cancer cells. Conclusion:2types of pancreatic cancer cells explained inhibited by 2-Deoxyglucose. Probably the mechanism lies in the apoptosis induces through caspase 3, and that may has a certain effect on anticancer chemotherapy or neoadjuvant chemotherapy in the next period.%目的:探讨糖酵解抑制剂2-脱氧葡萄糖(2-DG)联合广谱抗肿瘤药物顺铂对不同种系胰腺癌细胞系在不同情况下的体外抑制作用以及其机制.方法:使用2-DG以及联合顺铂对2种胰腺癌细胞系(Mia,Panc-1)实施生长干预,进行2-DG对不同细胞系抑制作用的MTT试验,及对细胞系进行凋亡的相关检测.结果:2-DG对不同种系的胰腺癌细胞抑制以及凋亡作用效果不同,但都呈时间反应性增加;2-DG联合顺铂较单药作用对细胞的抑制作用更为明显.结论:不同种系胰腺癌细胞系对2-DG都有一定的时间依赖性抑制作用,机制可能为2-DG对细胞凋亡的促进作用,2-DG在体外实验中体现一定的抗肿瘤活性以及联合化疗的活性,可能在胰腺癌的辅助化疗以及新辅助化疗等治疗中起到一定的作用.
