目的:探讨宫颈癌人乳头瘤病毒16型(HPVl6)E6和E7转化基因序列多态性,及其与宫颈癌临床病理特征的相关性。方法:抽取77例宫颈癌组织中筛选出51例HPVl6阳性标本,扩增E6及E7基因,PCR产物进行序列测定。应用DNA Star生物软件进行核苷酸和氨基酸序列的分析。结果:宫颈癌组织中HPVl6 E6共测得6个突变位点,均为错义突变。宫颈癌组织中 HPVl6 E7共测得4个突变位点,2个为错义突变。结论:HPVl6 E6基因 T178G/A突变与宫颈癌的发生发展无关,HPVl6 E7基因A647G突变与宫颈癌的分化程度呈负相关,可以作为评估官颈癌恶性程度的指标。%Objective :To study the human papilloma virus type 16 ( HPVl6 ) E6 and E7 transformed gene sequence polymorphism .Methods:Screening of 51 cases of HPVl6 positive samples were extracted in 77 cases of cervical carcinomas ,E6 gene and E7 gene amplification ,sequencing PCR products .Results :Cervical Cancer HPVl6 E6 mutations were measured six sites were missense mutations .Cervical cancer HPVl6 E7 were measured four mu-tations ,two missense mutations .Conclusion :the HPVl6 E6 gene T178G /A mutations in the pathogenesis and de-velopment of cervical cancer ,HPVl6 E7 gene A647G mutation was negatively related to the degree of differentiation and cervical cancer ,and can be used to assess the degree of malignancy of cervical cancer index .
